人胚胎干细胞向成纤维细胞分化方法的专家共识 被引量:4

摘要 人胚胎干细胞(ESC)是一类从囊胚期的人胚胎内细胞团中分离所得的,具有体外自我更新稳定性并维持正常核型和发育多能性,甚至参与整个个体发育的高度未分化干细胞[1]。利用人ESC或由其分化而成的人体细胞、组织或器官来测试各种生物医用材料、食品及药物对人体的毒理特性或药效,较运用动物或永生化细胞更能反映人体真实的状况。因此,人ESC及其分化的细胞有望发展成为一种崭新的生物安全性检测和药物筛选模型[2]。目前多数研究仍用永生化细胞系或原代培养细胞进行生物材料的安全性评价[3-4],分别存在核型异常或批次差异大等缺点。人ESC来源的成纤维细胞( ES-F )具有来源稳定,核型正常以及容易标准化等优点[5-6],有望成为生物安全性评价体系的新模型[7]。规范人ESC向成纤维细胞分化方法是保证生物安全性检测评价模型可靠性的关键。为此,科技部国际科技合作重点项目“创建基于人胚干细胞的预测健康安全新体系”的项目组专家经反复讨论,制定了“人胚胎干细胞向成纤维细胞分化方法的专家共识”(以下简称“共识”),旨在规范人ES-F的分化方法,提高人ES-F的纯度,保证分化方法的可重复性,使其更系统、规范和有效地应用于临床和生物安全性评价体系中。
