摘要
主要探讨了甲壳素纯度的测定方法。先用盐酸将甲壳素水解 ,然后将得到的水解产物———氨基葡萄糖盐酸盐 (GAH)进行衍生化 ,再用毛细管气相色谱法 (cGC法 )测定甲壳素的衍生物 ,并和比色法进行了比较。结果表明 :cGC法是一种灵敏的测定甲壳素纯度的方法 ,而比色法所得结果比期望值低。
A method for determining the purity of chitin was mainly studied. First, a given amount of chitin was hydrolyzed in 8 mol/L hydrochloride solution at 110℃ for 4 hours, and then the resulting glucosamine hydrochloride was reacted with N,O\|bis(trimethylsilyl) trifluoroacetamide and 1% trimethylchlorosilane in the hydroxylamine solution of pyridine at 70℃ for 20 minutes, and its derivative was analyzed by gas chromatography with a capillary column. The gas chromatography method was also compared with a colorimetric method for determining chitin. The result indicated that the gas chromatography was more sensitive and accurate than the colorimetry.
