摘要
【目的】研究亚低温对大鼠急性脑出血(intracerebral hemorrhage ,ICH)后脑组织含水量及核转录因子(nuclear factor‐κB ,NF‐κB)和细胞间黏附分子1(Intercellular adhesion molecule‐1,ICAM‐1)表达的影响。【方法】将50只健康雄性SD大鼠,随机(随机数字法)分为假手术组(SO)、常温组(NT )、亚低温组1 h组(HT1)、亚低温组2 h组(HT2)和亚低温组4 h组(HT4),每组10只大鼠,采用大鼠基底节注入自体不凝血法制作脑出血模型。SO组和NT组大鼠控制体温在(37.0±0.3)℃;亚低温组在脑出血后迅速降温至(33.0±0.4)℃并分别保持1 h、2 h和4 h。检测各组大鼠脑组织含水量,凝胶电泳迁移率实验(EMSA )检测 NF‐κB活性;Western blot 检测ICAM‐1蛋白的表达。【结果】与SO组相比较,NT组大鼠脑组织含水量显著升高,NF‐κB活性及ICAM‐1蛋白表达均显著降低,其差异有统计学意义( P <0.05);NT 组与 HT1、HT2和 HT4组大鼠脑含水量、NF‐κB活性及ICAM‐1蛋白表达比较均有显著性差异( P <0.05);HT2组和 HT4组相比较,大鼠脑含水量、NF‐κB活性及ICAM‐1蛋白表达比较均无统计学意义( P >0.05)。【结论】早期亚低温治疗可抑制大鼠脑出血后NF‐κB活性和IC A M‐1蛋白的表达,从而减轻脑水肿。
[Objective] To explore the effect of mild hypothermia on water content ,nuclear factor‐B(NF‐κB) and intercellular adhesion molecule‐1 (ICAM‐1 ) in brain tissue of rats after intracerebral hemorrhage (ICH ) .[Methods] Totally 50 healthy male Sprague‐Dawley(SD) rats were randomly(random number method) divided in‐to sham‐operated group(SO group) ,normothermic group(NT group) ,1h‐mild‐hypothermia group(HT1 group) , 2h‐mild‐hypothermia group(HT2 group) and 4h‐mild‐hypothermia group(HT4 group) with 10 rats in each group . ICH model was established by stereotactic infusion of autologous blood into caudate nucleus .Rats in NT group and SO group were kept at (37 .0 ± 0 .3)℃ of body temperature .Rats in HT groups after ICH were kept at (33 .0 ± 0 .4) ℃ of body temperature for 1 ,2 and 4h ,respectively .Water content in brain tissue of rats in each group was measured .NF‐κB activity was detected by electrophoretic mobility shift assay (EMSA) .The expression of ICAM‐1 was detected by Western blot .[Results] Compared with SO group ,brain water content and the expression of NF‐κB and ICAM‐1 in NT group were markedly increased ,and there was significant difference( P〈0 .05) .Brain water content and the expression of NF‐κB and ICAM‐1 in HT1 ,HT2 and HT4 groups were significantly lower than those in NT group( P〈0 .05) .There was no significant difference in brain water content and the expression of NF‐κB and ICAM‐1 between HT2 group and HT4 group( P〉0 .05) .[Conclusion]Early mild hypothermia can inhibit the expression of NF‐κB and ICAM‐1 in rats after intracerebral hemorrhage ,and alleviate brain edema .
