聚合物包衣在固体药物制剂尤其是固体缓控释递送系统中应用极为普遍,然而使用单一包衣辅料很难达到理想的药物释放曲线。混合使用包衣辅料在克服上述缺陷的同时显示出诸多优势:1便于调节目标药物释放行为和药物释放机制。2提高聚合物成膜性、衣膜机械性和长期稳定性。3设计具有胃肠道特定部位给药的制剂(如:结肠靶向)。本文概述了功能性包衣辅料的混合使用在药物控释递送系统中的应用进展,重点介绍了混合使用对药物释放行为的影响和相应的药物释放机制。
Polymeric film coatings are frequently used to control drug release from solid pharmaceutical dosage forms, but a single polymer is often difficult to obtain a desired release profile. The use of polymer blends as coating materials can overcome this pitfall by several major advantages, including : ① facilitating adjustment of desired drug release patterns, mechanical properties and drug release mechanisms, ② improving film formation and storage stability, and ③the possibility to develop novel strategies for site specific drug delivery within the gastroin- testinal tract (e. g. , colon targeting). This article gives an overview on the current state of the art of the use of polymer blends as coating materials for controlled drug delivery, explaining the major advantages. Special emphasis is laid on the effects of the polymer blend ratio on the drug release and the underlying drug release mechanisms.
