摘要
目的优选骆驼蓬籽中脂肪酸定量分析的甲酯化条件。方法以5种脂肪酸(油酸、亚油酸、亚麻酸、棕榈酸、花生酸)的总量、甲酯化峰个数及百分峰面积的综合评分为优选指标,采用正交试验分别对4种甲酯化过程进行条件优选,并通过横向比较确定合适的脂肪酸甲酯化条件。结果 1%H2SO4-甲醇回流法更适合用于骆驼蓬籽中脂肪酸的甲脂化,优选条件为醇油体积比4∶1,催化剂与油的体积比2∶1,反应温度为80℃,回流时间为30 min。结论验证试验表明优选出的甲酯化工艺稳定性好、反应时间短,可作为气相色谱法的前处理方法,用于骆驼蓬籽中脂肪酸含量测定。
AIM To select the conditions of methyl esterification for the quantitative analysis of fatty acids from the seeds of Peganum harmala. METHODS A comprehensive scoring was taken,based on the total content of five fatty acids( oleic acid,linoleic acid,linolenic acid,palmitic acid,and arachic acid),the peak numbers and area percentage of peaks,with the orthogonal design employed to compare and figure out the optimal conditions of four different methyl esterifications for fatty acids. RESULTS 1% sulfuric acid-methanol reflux method was determined as a satisfying methyl esterification,which consisted of 4 ∶ 1 volume ratio of methanol to oil,2 ∶ 1volume ratio of catalyst to oil,80 ℃ reaction temperature,and 30 min reaction time. CONCLUSION The validation tests indicate that the optimum process is stable and speedy,and it lays a foundation for gas chromatography of fatty acids in P. harmala.
