摘要
目的利用HPLC法对草原老鹳草中莽草酸和没食子酸进行定量测定,建立草原老鹳草定量分析的质量控制标准。方法 Agilent TC-C18色谱柱(4.6 mm×250 mm,5μm),以乙腈-1%磷酸溶液(5∶95)为流动相,体积流量为0.6 m L/min,检测波长为211 nm,柱温25℃。结果莽草酸、没食子酸的线性范围分别为0.196-1.372μg(r=0.999 5)、0.270-1.890μg(r=0.999 9),平均回收率为99.7%(RSD 1.7%)、101.5%(RSD 1.6%)。结论 9批药材中莽草酸平均质量分数为1.937 9 mg/g,没食子酸平均质量分数为2.438 7 mg/g,可作为草原老鹳草药材的质量控制方法。
AIM To establish an HPLC method for the determination of shikimic acid and gallic acid in Geranium pratense L. METHODS The analytical column was Agilent TC-C18column( 4. 6 mm × 250 mm,5 μm)with acetonitrile-1% aqueous acetic acid( 5 ∶ 95) as mobile phase. The flow rate of 0. 6 m L / min,the UV detection was set at 211 nm and the column temperature was at 25 ℃. RESULTS The linear ranges were 0. 196 ~1. 372 μg for shikimic acid( r = 0. 999 5) and 0. 270 ~ 1. 890 μg for gallic acid( r = 0. 999 9),respectively.The average recoveries of shikimic acid and gallic acid were 99. 7%( RSD 1. 7%) and 101. 5%( RSD 1. 6%),respectively. CONCLUSION The nine batches of herbs that the average content of shikimic acid is 1. 937 9mg / g and the average content of gallic acid is 2. 438 7 mg / g. The method could be used for quality control for this herb.
