反相高效液相色谱法测定纤体产品中左旋肉碱的含量 被引量:5

摘要 目的建立了反相高效液相色谱(RP-HPLC)法测定纤体产品中左旋肉碱的含量。方法采用L-丙氨酸-β-萘胺(L-Ala-β-NA)作为柱前衍生化试剂,色谱柱为Agilent ZORBAX SB-C18(4.6 mm×250 mm,5μm),以乙腈-磷酸二氢钠缓冲液(4∶96)为流动相,检测波长为244 nm,流速为l mL·min-1,柱温30℃。结果左旋肉碱与右旋肉碱色谱峰达到基线分离,左旋肉碱在11.565~115.65 ng与峰面积线性关系良好,r为0.999 0,加样回收率为95.6%~100.0%。结论该方法经济可行,操作较简便,结果准确,重复性好,可用于纤体产品中左旋肉碱含量的测定。 Objective To establish a method for determination of L-carnitine in slimming products by RP-HPLC. Methods The samples were derivatized with chiral compound L-alanine-β-naphthylamide and the separarion was performed on an Agilent ZORBAX SB-C18 column (4.6 mm×250 mm, 5 μm),the mobile phase consisted of acetonitrile-tetrahydrofuran-phosphate buffer solution (4 : 96). The flow rate was 1 mL · min- 1. The detection wavelength was 244 nm and the column temperature was 30 ℃ . Results The resolution for the optical isomer of carnitine was good. The linear range of L-camitine was 11.565 - 115.65 ng (r= 0.999 0). The recoveries ranged from 95.6% to 100.0%. Conclusion The improved method is economically viable, simple, accurate and reliable, which can be applied to the quality control of L-camitine in slimming products.
