透明质酸刺激因子对增生性瘢痕成纤维细胞透明质酸合成的影响

摘要 目的 了解兔源性透明质酸刺激因子 (HASF)对体外单层及三维培养条件下增生性瘢痕成纤维细胞 (FB)透明质酸 (HA)合成的影响 .方法 从胎兔羊水及血清中提纯HASF分别作用于单层及三维培养的 FB细胞 ,用放免法测定上清液中 HA浓度 .结果 在单层培养条件下 ,随着 HASF剂量由 0 .1m g· L- 1增至 0 .1g· L- 1 ,HA浓度也由 (495±37) mg· L- 1 增至 (914± 75 ) mg· L- 1 ,在一定范围内呈剂量依赖性 (P<0 .0 1) .继续增加 HASF剂量 ,HA浓度并不成比例增加 ,呈显出饱和性 .随 HASF作用时间由 3d延长至 7d,HA浓度也呈时间依赖性的增加 (P<0 .0 1) .在三维培养条件下 ,随 HASF剂量的增加 ,HA浓度也由 (90± 2 5 ) mg· L- 1增至 (2 93± 6 3) mg· L- 1 ,也呈剂量依赖性 (P<0 .0 1) ,且明显低于单层培养条件下 HA的浓度 (P<0 .0 1) .结论  HASF对单层及三维培养的 FB有明显的促 HA合成作用 ,并呈剂量和时间依赖性 . AIM To investigate the effect of hyaluronic acid stimulating factor (HASF) on the hyaluronic acid (HA) syn thesis of hypertrophic scar derived fibroblasts cultured three dimensionally in the monolayer. METHODS HASF, isolated from the fetal rabbit sera and amniotic fluid, was added to the monolayer and three dimensional fibroblast culture systems. The concentration of HA in the supernatant of the fibroblast culture systems was measured using radioimmunoassay. RESULTS Concentration of HA in the monolayer system increased from (495±37) mg·L -1 to (914±75) mg·L -1 in a dose dependent manner with the increase of HASF from 0.1 mg·L -1 to 0.1 g·L -1 ( P <0.01). The concentration of HA reached the maximam when HASF at 0.1 g·L -1 and stopped increasing in proportion to the concentration of HASF. The concentration of HA increased in a time dependent manner in the presence of HASF between 3rd and 7th. In three dimensional culture system, the concentration of HA increased from (90±25) mg·L -1 to (293±63) mg·L -1 in a dose dependent manner ( P <0.01). CONCLUSION HA synthesis of hypertrophic scar derived fibroblasts can be stimulated by HASF both in monolayer and three dimensional fibroblast culture systems in a dose and time dependent manner.
