大黄素调节Sirt1表达改善脂肪细胞糖代谢的研究 被引量:4

摘要 目的研究大黄素对脂肪细胞葡萄糖代谢的影响及其机制。方法以分化成熟的脂肪细胞为研究对象,检测10μmol/L的大黄素对脂肪细胞葡萄糖代谢的影响。以流式细胞术检测脂肪细胞对2-NBDG荧光葡萄糖类似物的吸收;以免疫荧光-激光共聚焦技术检测葡萄糖转运蛋白4在细胞内的定位和表达;以qPCR检测Sirt1基因的mRNA的表达;以Western Blot检测Sirt1蛋白的表达。结果流式细胞术检测葡萄糖吸收实验结果表明,大黄素可显著促进成熟脂肪细胞对葡萄糖的吸收。经10μmol/L大黄素处理脂肪细胞3h后,葡萄糖的吸收量平均增加了46.01%(P<0.05)。免疫荧光显示经大黄素处理的脂肪细胞的葡萄糖转运蛋白4(GLUT4)显著从细胞内膜泡转位至细胞膜表面(P<0.01);同时,大黄素使Sirt1的基因表达与蛋白表达都显著增加(P<0.05),这可能是大黄素改善脂肪细胞葡萄糖代谢的机制之一。结论大黄素可能通过调节Sirt1表达改善脂肪细胞的葡萄糖代谢。 OBJECTIVE To study the influence of emodin on the glucose metabolism and its possible mechanism.METHODS The effects of 10μm emodin on adipocyte glucose metabolism with the differentiated adipocytes as research object were studied and the intracellular location and expression,Sirt1 genetic mRNA expression and the expression of Sirt1 protein by immunofluorescence-confocal laser technology,qPCR and Western Blot were also detected.RESULTS Experimental results by FCM (flow cytometry) showed that Emodin increased the uptake of glucose in adipocytes to 46.01% compared to the control group.It showed that emodin may promote the translocation of GLUT4 from cytoplasm to cell membrane significantly.Compared with the control group,the expression of Sirt1 mRNA and protein of adipocytes treated with emodin was significantly increased respectively.This may be one of the mechanisms of emodin in improving glucose metabolism of adipocytes.CONCLUSION Emodin may improve glucose metabolism by up-regulating the expression of Sirt1.
