摘要
以甲醇与丙二腈为主要原料,在无水甲苯的氯化氢体系中发生亲核加成反应合成1,3-二甲氧基丙二亚胺盐酸盐;碱中和后与单腈胺反应生成3-氨基-3-甲氧基-N-腈基-2-丙咪;甲苯中回流环合生成2-氨基-4,6-二甲氧基嘧啶。采用单因素法考察了反应溶剂、反应时间、反应温度等对产品收率的影响。得到了合成目标产物的最佳条件,在此条件下目标产品的最终收率为75%(以丙二腈记)。
2-amino-4,6-dimethoxy pyrimidine was prepared by using methyol and malononitrile as main materials in the following steps. 1,3 -dimethoxy propyleneimine hydrochloride was firstly made in anhydrous toluene,then reacted with cyanamide to give 3 -amino -3 -methoxy -N - nitrile propionamidine,finally cyclized in toluene to give the target product. Different reaction conditions such as reaction solvent,time and temperature were discussed. The best yields can reach up to 75% at the optional conditions.
