摘要
目的建立胶原诱导性关节炎(collagen induced arthritis,CIA)大鼠滑膜成纤维细胞的培养方法并观察细胞生物学特点。方法收集CIA和正常大鼠的膝关节滑膜组织,采用组织块贴壁法培养滑膜成纤维细胞,观察细胞游出、生长情况;HE染色观察细胞形态;免疫细胞化学染色鉴定细胞纯度;Masson染色观察细胞分泌胶原蛋白情况;CCK-8法检测细胞增殖情况。结果倒置显微镜及HE染色观察细胞形态及生长特征符合滑膜成纤维细胞,且98%以上的细胞表达Vimentin蛋白。模型组滑膜成纤维细胞较正常组细胞游出时间早且细胞量多;模型组细胞分泌胶原蛋白能力及细胞增殖能力较正常组强。结论本研究确立了CIA大鼠滑膜成纤维细胞原代培养体系,并且证实CIA大鼠滑膜成纤维细胞增殖与分泌功能较正常滑膜成纤维细胞强。
To establish methods for primary culture of CIA rat synovioblast and to observe the biological characteristics of the cells, the knee synovial tissues of the CIA and normal rats were cultured and the growth of the synovioblast was observed. Furthermore, the morphology of the cells was observed by HE staining, the purity of the cell was identified by immunocytochemistry staining, the ability to secrete collagen of the cells was observed by Masson staining, and the cell proliferation was measured by CCK-8 assay. Data showed that the morphology and biological characteristics of the cultured cells were coincided with synovioblast, and more than 98% cells expressed vimentin. Compared with the normal synovioblast, the synovioblast of CIA rats swam out earlier and more cells swam out, secreted more collagen and with higher proliferative ability. This study suggests that the method for primary culture of CIA rat synovioblast is successfully established and proves that the proliferative and secretory ability of the CIA rat synovioblast was higher than that of normal synovioblast.
