摘要
目的:研究连翘苷对衰老模型小鼠的抗衰老作用。方法:小鼠颈部皮下注射D-半乳糖(150 mg/kg),每天1次,连续42 d以复制小鼠衰老模型。50只KM小鼠随机均分为正常对照(等容甲基纤维基钠)组、模型(等容甲基纤维基钠)组与连翘苷高、中、低剂量(45、15、5 mg/kg)组,复制模型同时灌胃给药,每天1次,连续42 d。测定小鼠增质量率与脾脏、心脏、肾脏、肝脏指数,测定小鼠血清、肝组织超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)、总抗氧化能力(T-AOC)水平,测定小鼠脑组织MDA、B型单胺氧化酶(MAO-B)、SOD、T-AOC水平。结果:各组间心脏、肾脏、肝脏指数差异无统计学意义(P>0.05)。与正常对照组比较,模型组小鼠增质量率降低,脾脏指数降低;小鼠血清、肝组织SOD、GSH-Px、T-AOC活性减弱,MDA含量增加;小鼠脑组织MDA含量增加,MAO-B活性增强、SOD、T-AOC活性减弱,差异均具有统计学意义(P<0.01或P<0.05)。与模型组比较,连翘苷高、中、低剂量组小鼠增质量率升高,脾脏指数升高;连翘苷高、中剂量组小鼠血清、肝组织SOD、GSH-Px、T-AOC活性增强,MDA含量减少;连翘苷高、中、低剂量组小鼠脑组织MAO-B活性减弱;连翘苷高、中剂量组小鼠脑组织MDA含量减少,差异均具有统计学意义(P<0.01或P<0.05)。结论:15、45 mg/kg剂量下,连翘苷对衰老模型小鼠具有较好的抗衰老作用。
OBJECTIVE: To study the anti-aging effects of phillyrin on aging model mice. METHODS: The aging model was induced by subcutaneous injection of D-galactose (150 mg/kg) on cervical region once a day for consecutive 42 d. 50 KM mice were randomly divided into normal control group (constant volume of CMC-Na), model group (constant volume of CMC-Na), phillyrin high-dose, medium-dose and low-dose groups (45, 15 and 5 mg/kg). They were given relevant medicines intragastrically once a day for consecutive 42 h. The rate of weight gain, the index of spleen, heart, kidney and liver were determined in mice; the levels of SOD, GSH-Px, MDA and T-AOC in serum and liver tissue of mice were all detected; the levels of MDA, MAO-B, SOD and T-AOC were detected in brain tissue of mice. RESULTS: There was no statistical significant difference in the index of heart, kidney and liver (P〉0.05). Compared with normal control group, the rate of weight gain, the index of spleen, the activi- ties of SOD, GSH-Px and T-AOC in serum and liver tissue were decreased in model group, while the content of MDA was in- creased; MDA content and MAO-B activity in brain tissue of mice were increased, while SOD and T-AOC activities were de- creased; there were statistical significant difference (P〈0.01 or P〈0.05). Compared with model group, the rate of weight gain and the index of spleen were increased in phillyrin high-dose, medium-dose and low-dose groups; SOD, GSH-Px and T-AOC activ- ities in serum and liver tissue were increased in phillyrin high-dose and medium-dose group, while MDA content was decreased; MAO-B activity of brain tissue was decreased in phillyrin high-dose, medium-dose and low-dose groups; MDA content of brain tis- sue was decreased in phillyrin high-dose and medium-dose groups; there were statistical significant difference (P〈0.01 or P〈0.05). CONCLUSIONS: Phillyrin (15, 45 mg/kg) possesses satisfying function of anti-aging on aging model mice.
