摘要
目的:研究心得宁口服液对慢性心衰模型大鼠心脏动力学和血清B型脑钠肽(BNP)、心肌肌酐蛋白(c Tn)Ⅰ、乳酸脱氢酶(LDH)水平的影响。方法:通过腹主动脉缩窄法复制大鼠慢性心衰模型。72只Wistar雄性大鼠随机均分为假手术(等容生理氯化钠溶液)组、模型(等容生理氯化钠溶液)组、芪苈强心胶囊(0.6 g/kg)组与心得宁口服液高、中、低剂量(20、10、5 ml/kg)组,灌胃给药,每天1次,连续6周。检测大鼠心脏动力学指标,大鼠血清BNP、c TnⅠ含量与LDH活性。结果:与假手术组比较,模型组大鼠心率(HR)减慢,左室收缩内压(Lvsp)、左室内压最大变化速率(±dp/dtmax)降低,左室舒张末压(Lvedp)升高,BNP、c TnⅠ含量增加,LDH活性增强,差异均具有统计学意义(P<0.01或P<0.05)。与模型组比较,心得宁口服液高、中、低剂量组大鼠Lvsp升高,LDH活性减弱;中、低剂量组大鼠血清BNP含量减少;高剂量组大鼠血清c TnⅠ含量减少;中剂量组大鼠HR增加,差异均具有统计学意义(P<0.01或P<0.05)。结论:心得宁口服液可明显改善慢性心衰模型大鼠心脏动力学指标与血清生化指标。
OBJECTIVE: To study the effects of Practolol oral solution on cardiac dynamics and serum levels of BNP, cTn I and LDH in rats with chronic heart failure. METHODS: Chronic heart failure model was induced by abdominal aorta coarctation. 72 Wistar male rats were randomly divided into sham operation group (constant volume of sodium chloride physiological solution), model group (constant volume of sodium chloride physiological solution), Qili qiangxin capsule group (0.6 g/kg), Practolol oral solution high-dose, medium-dose and low-dose groups (20, 10, 5 ml/kg). They were given relevant medicine intragastrically once a day for consecutive 6 weeks. The cardiac dynamics indicators of rats were determined. The serum contents of cTn I and BNP, LDH activity were all determined in rats. RESULTS: Compared with sham operation group, in model group, HR was decreased, Lvedp, the contents of BNP and cTn I , LDH activity were increased, while Lvsp and dp/dtwas decreased; there were statisti- cal significant difference (P〈0.01 or P〈0.05). Compared with model group, Lvsp levels of Practolol oral solution high-dose, medium-dose and low-dose groups were increased, while LDH activities were decreased; the serum contents of BNP was decreased in Practolol oral solution medium-dose and low-dose groups; the contents of cTn I was decreased in Practolol oral solution high-dose group; HR of Praetolol oral solution medium-dose group was increased; there were statistical significant difference (P〈0.01 or P〈 0.05). CONCLUSIONS: Practolol oral solution can significantly improve cardiac dynamics and serum biochemical indicator in rats with chronic heart failure.
