七氟烷对内毒素致急性肺损伤鼠中性粒细胞趋化因子-1和细胞间黏附分子-1的影响 被引量:3

摘要 目的观察七氟烷对内毒素脂多糖(LPS)致急性肺损伤(ALI)大鼠肺组织中性粒细胞趋化因子-1(CINC-1)和细胞间黏附分子-1(ICAM-1)的影响。方法 Wistar大鼠40只,随机分为4组(n=10),对照组股静脉注射生理盐水1.5 m L后机械通气4 h;LPS组股静脉注射LPS 5 mg/kg后机械通气4 h;1.0、1.5肺泡最低有效浓度(MAC)七氟烷组:股静脉注射LPS 5 mg/kg后机械通气分别吸入1.0和1.5 MAC七氟烷4 h。实验结束时心脏放血处死大鼠,取左肺HE染色后普通光学显微镜观察肺组织的损伤程度及免疫组化测定CINC-1、ICAM-1,右肺行支气管肺泡灌洗,收集支气管肺泡灌洗液(BALF)1 m L进行细胞计数,余灌洗液离心后取上清液ELISA法测定CINC-1及ICAM-1。结果 LPS组BALF内细胞总数和中性粒细胞比例明显高于对照组(P<0.01),七氟烷组细胞总数及中性粒细胞比例明显低于LPS组(P<0.05),但高于对照组(P<0.05);LPS组BALF上清液中CINC-1、ICAM-1浓度和肺组织内免疫组化阳性表达明显高于对照组(P<0.01);七氟烷组低于LPS组,高于对照组(P<0.05);七氟烷组肺组织损伤评分低于LPS组,高于对照组(P<0.05)。结论七氟烷可以降低LPS所致ALI大鼠BALF内细胞总数及CINC-1、ICAM-1浓度,以及肺组织内CINC-1、ICAM-1蛋白表达,减轻肺组织损伤。
