摘要
以L-酪氨酸、乙酰氯为主要原料合成了N-乙酰-L-酪氨酸,再和乙醇与氯化亚砜反应得到的氯化亚硫酸乙酯反应得到N-乙酰-L-酪氨酸乙酯,产物结构经IR、1H NMR确证.单因素试验和正交试验结果表明,N-乙酰-L-酪氨酸乙酯合成的最佳工艺条件为:n(L-酪氨酸)∶n(氯化亚砜)∶n(乙醇)=1∶1.3∶10,加热温度75℃,加热时间2 h.该工艺具有原料价廉易得、反应周期短、产物易分离的特点,适于工业化大规模生产.
We prepared N-acetyl-L-tyrosine from L-tyrosine and acetyl chloride,then synthesized N-Acetyl -L-tyrosine ethyl ester from N-acetyl-L-tyrosine and ethyl sulfite chloride which was the product of ethanol and thionyl chloride, and finally confirmed the structure of the product by IR and 1H NMR. Through single factor and orthogonal tests,we determined the optimum synthesis conditions as follows: the molar ratio of L-tyrosine to SOC12 to ethanol 1:1.3:10, reaction temperature 75 ℃ and reaction time 2 hours. The synthesis process had the characteristics of cheap and obtainable starting materials, short reaction period, and simple product separation, so that the process is suitable for industrial production.
