摘要
目的观察温清饮对HaCaT细胞增殖和分化的影响。方法用细胞体外培养技术检测温清饮对HaCa T细胞增殖和分化的影响。结果温清饮对HaCaT细胞的增殖具有抑制作用,最小抑制浓度为5 mg/m L,呈现剂量依赖的关系。温清饮影响HaCaT细胞FLG基因的表达,经温清饮处理48 h后的HaCaT细胞其FLG mRNA的表达水平明显高于对照组。结论温清饮对HaCaT细胞的增殖具有抑制作用并上调HaCaT细胞FLG mRNA的表达。
Objective：To investigate the effects of Wenqingyin on the proliferation and differentiation of HaCaT cells.Methods：Cultured HaCaT cells in vitro were used to test the effects of Wenqingyin in various durations on the Proliferation and Differentiation of HaCaT Cells.The cell proliferation was assessed with CCK-8 method,the changes of filaggrin （FLG） mRNA expressions by semi-quantitative reverse transcription PCR.Results：The proliferation of HaCaT cells was inhibited by the treatment with Wenqingyin with the minimum inhibiting concentration 5 mg/mL,and the inhibition rate rose with the increase of the dosage,which was dose-dependent.With the use of Wenqingyin,the mRNA expression of FLG was higher than that of the control group.Conclusion：Wenqingyin can inhibit the proliferation and induce the differentiation of keratinocytes.
