摘要
细胞内p H在许多细胞生物学过程中发挥着至关重要的作用,异常的p H变化会导致细胞功能紊乱、生长和分裂的突变,还可能引发癌症和阿尔茨海默综合症等疾病。因此,研究细胞内p H的变化具有非常重要的理论和实践意义。荧光分析法具有操作简便、灵敏度高、选择性好、实时检测以及对生物体损伤小等优点而得到了广泛地研究和应用。双光子荧光探针技术相对于单光子荧光技术具有长波吸收、短波发射、高度的三维空间选择性、大的穿透深度、避免荧光漂白和光致毒以及降低组织自发荧光干扰等特点,在生命科学领域显示出了广阔的应用前景。介绍了有机单光子和双光子p H荧光探针的研究现状,同时对有机p H荧光探针未来的发展趋势进行了展望。
Intracellular p H plays an important role in many biological processes,which could induce cell dysfunction,growth or division and may cause diseases such as cancer and alzheimer's syndrome. Therefore,the research of the intracellular p H's change is significantly meaningful in theory and practice. Fluorescence methods have been extensively studied and applied due to simple operation,high sensitivity and selectivity,real-time detection and little damage to organisms. The two-photon fluorescent probe technique has shown much broader application prospect in the field of life sciences compared to the one-photon fluorescent probe technique due to many features,such as absorbing light of longer wavelength with lower energy but emitting shorter wavelength,high three-dimensional spatial selectivity,large penetration depth,avoidance of photodamage and photobleaching and lower tissue auto-fluorescence,etc. In this work,the progress of organic one-photon and two-photon p H fluorescent probes were introduced,as well as the developing trends of organic p H fluorescent probes in the future were prospected.
