摘要
目的:通过检测非综合征性唇腭裂(NSCL/P)患者与正常人尿液中代谢物的变化,探讨基于核磁共振(NMR)的代谢组学在此症的临床应用中的可行性。方法:分别收集12个NSCL/P患者和12个正常人对照组的尿液样品。采用NMR分析NSCL/P患者尿液代谢特征,测定1 H-NMR图谱,进而用主成分分析法(PCA)处理NMR数据,研究NSCL/P组和正常对照组尿液中的代谢物差异,获得NSCL/P患者的特异性代谢标志物。结果:NSCL/P组和正常对照组间的尿液代谢产物存在明显差别。结论:采用基于NMR的代谢组学技术能较好地反映NSCL/P患者尿液的代谢变化,获得相应的特征代谢物,在未来的临床应用中具有广阔的前景。
Objective: To detect the metabolites in urine of non-syndromic cleft lip and/or palate(NSCL/P) patients and to explore the feasibility of employing nuclear magnetic resonance(NMR)-based metabonomics in clinical practice.Method: Urine samples were collected from 12 NSCL/P patients group and 12 healthy control group. Urine metabolomic profiles were assessed by 1 H-NMR spectroscopy and principal component analysis(PCA). Discriminating metabolites were identified, and metabonomic differences of urine were examined between NSCL/P patients group and the control group. Result: The urine metabolic profiling in NSCL/P patients group was remarkably distinct from that of healthy control group. Conclusion: The application of NMR-based metabonomics techniques reflected urine metabolic changes of patients with NSCL/P,indicating the great potential of metabonomics in clinic diagnosis and screening.
