加味清营颗粒对急性肺损伤大鼠肺组织AQP-1表达的影响 被引量:3

摘要 目的探讨加味清营颗粒对急性肺损伤大鼠肺组织水通道蛋白-1(AQP-1)表达的影响。方法清洁级成年雄性Wistar大鼠144只,随机分为空白对照组、模型对照组、加味清营汤低剂量组、加味清营汤中剂量组、加味清营汤高剂量组、地塞米松组6组,每组24只。除空白对照组外,其余各组采用内毒素制备急性肺损伤大鼠模型。于造模后1 h、3 h、5 h时间点分别处死大鼠,采用免疫组化法观察大鼠肺组织AQP-1表达。结果加味清营汤高、中、低剂量组与模型组在AQP-1表达上比较差异有统计学意义;加味清营汤高剂量组与地塞米松组在AQP-1表达上比较差异无统计学意义;加味清营汤高、中、低剂量组之间在AQP-1表达上呈现正相关量效关系;AQP-1表达随急性肺损伤时间呈负相关关系。结论加味清营颗粒能有效提高急性肺损伤大鼠肺组织AQP-1的阳性表达,纠正水转运紊乱,从而起到肺组织保护作用。 Objective To explore the impacts of jiawei qingying granules on AQP- 1 expression in lung tissue of the rats with acute lung injury. Methods One hundred and forty- four clear- grade,male adult Wistar rats were randomized into a blank control group,a model control group,a low- dose jiawei qingying granules group( low dose group),a middle- dose jiawei qingying granules group( middle dose group),a high- dose jiawei qingying granules group( high dose group) and a dexamethasone group,24 cases in each one. Except the blank control group,the endotoxin was used to prepare acute lung injury rat models in the rest groups. In 1h,3h and 5h after modeling,the rats were sacrificed. IMC was adopted to observe AQP- 1 expression in lung tissue. Results AQP- 1 expression in each of jiawei qingying granules groups was different significantly as compared with that in the model group. The positive dose- effect correlation presented among jiawei qingying granules groups. The negative correlation presented between AQP- 1 expression and the duration of acute lung injury. Conclusion Jiawei qingying granules effectively improve AQP- 1 positive expression in lung tissue of the rats with acute lung injury and rectify water transportation disorder so as to protect lung tissue.
