摘要
为了研究黑果枸杞对运动性肾缺血再灌注大鼠血清TNF-α、IL-6、IL-10含量及肾组织ICAM-1表达的影响,将7周龄雄性Wistar大鼠75只随机分为4组,即C组(对照组)、M组(一般训练组)、OM组(过度训练组)和LOM组(黑果枸杞+过度训练组),M组、OM组和LOM组进行6周的游泳训练。LOM组以黑果枸杞灌胃,其他各组灌胃等量生理盐水。末次训练后24h检测血清TNF-α、IL-6、IL-10含量及肾组织ICAM-1表达。结果显示:血清TNF-α、IL-6含量及肾组织ICAM-1表达,OM组和LOM组高于C组(P<0.01)、LOM组低于OM组(P<0.05);血清IL-10含量,OM组(P<0.01)和LOM组(P<0.05)低于C组,LOM组高于OM组(P<0.05)。实验结果说明6周过度训练导致大鼠运动性肾缺血再灌注。黑果枸杞可能通过抑制促炎性细胞因子TNF-α、IL-6的分泌及肾组织ICAM-1表达和促进抑炎性炎症因子的IL-10的分泌,提高机体免疫能力,提高耐受各种应激刺激的能力,对过度训练导致的运动性肾缺血再灌注损伤有保护作用。
To study the protective effects of Lycium Ruthenicum Murr. on the exercise-related renal ischemia-reperfusion injury in rat serum TNF-a, IL-6, IL-10 and expression of renal tissue ICAM-1. 75 male Wistar rats at the age of 7-week-old were divided into 4 groups randomly in test., control group (C group, n= 12), general training group (M group, n= 12), overtraining group (OM group, n=24), Lycium Ruthenicum Murr.+overtraining group (LOM group, n=24). Rats in M, OM, LOM groups were assigned to go on 6 weeks of swimming training. Professional gavage were taken daily. The Rats in LOM group were given with 4. 48 g · kg^-1 Lycium Ruthenicum Murr. gavage at 5mL·kg^-1 and other groups were given the normal saline. 24 hours after the last training, we detect the content of serum IL-1β, TNF-α, IL- 6, IL-10 and the expression of renal tissue ICAM-1 by Enzyme-linked Immunospot Assay(Elisa)and immunohistochemistry. The content of serum TNF-α, serum IL-6 and expression of ICAM-1 in nephridial tissue in both OM group and LOM group were higher than C group (P〈0. 01). In the mean time, all these indexes in LOM group were lower than OM group (P〈0. 05). The content of serum IL-10 in OM group and in LOM group were lower than C group (OM, P〈0. 01;LOM,P〈0. 05),and the content of serum IL-10 in LOM group were higher than OM group (P%0. 05). The results showed that six weeks overtraining will led to exercise-related renal ischemia-reperfusion injury (ERRIRI) in rats. Lycium ruthenicum Murr. can inhibit the secretion of inflammatory cytokines including TNF-α and serum IL-6. In the mean time, Lycium ruthenicum Murr. can decrease the expression of ICAM-1 in nephridial tissue and increase the secretion in IL-10, then improve the immune function to tolerance the stimulation from outside environment. After supplement of Lycium ruthenicum Murr. can protect ERRIRI from overtraining.
