摘要
目的观察研究早期使用三七总皂苷对脑出血大鼠的神经功能缺损状况以及炎性反应递质的影响。方法将雄性SD大鼠随机分成假手术组、模型组、三七总皂苷组,采用胶原酶注射尾状核法制作脑出血模型,观察大鼠脑出血死亡率、神经功能缺损情况以及血肿周围白介素(IL)-6和基质金属蛋白酶(MMP)-9的表达水平。结果 3组大鼠在造模的过程中,均有死亡病例发生。其中,假手术组脑出血死亡率(脑出血死亡数/总死亡数)低于模型组和三七总皂苷组(均P<0.05),而模型组和三七总皂苷组脑出血死亡率差异不大(P>0.05)。术后24 h和术后第7日,模型组和三七总皂苷组的神经功能缺损评分均高于假手术组(P<0.05)。术后24 h比较,模型组和三七总皂苷组神经功能缺损评分无明显差异(P>0.05)。术后第7日比较,三七总皂苷组的神经功能评分及其降低的分值均明显优于模型组(P<0.05)。术后48 h,模型组以及三七总皂苷组血肿周围的IL-6和MMP-9的积分光密度均明显高于假手术组(均P<0.05)。而模型组与三七总皂苷组比较,血肿周围的IL-6和MMP-9的积分光密度差别不大(均P>0.05)。术后第7日,IL-6、MMP-9的积分光密度模型组与三七总皂苷组明显高于假手术组(均P<0.05),且三七总皂苷组的积分光密度均低于模型组(均P<0.05)。3组在术后第7日时,IL-6的表达均较48 h增高(均P<0.05),其中模型组升高最明显,三七总皂苷组和假手术升高都相对较缓;假手术组和三七总皂苷组在术后第7日时MMP-9的表达均较48 h降低(均P<0.05),而模型组较前升高(P<0.05)。结论三七总皂苷可以减轻大鼠神经功能缺损,抑制炎性因子IL-6、MMP-9的释放,且用药安全。
Objective: To observe the PNS on improving neurological deficit and the effects of inflammatory mediators in rat brain hemorrhage and to provide a theoretical basis for the early application of cerebral hemorrhage.Methods: Male SD rats were randomly divided into sham operation group,model group,the group of PNS. Collagenase injections caudate nucleus produced cerebral hemorrhage model was bleeding mortality,neurological deficit,the hematoma around the rat brain expression of IL-6 and MMP-9. Results: In the first seven days after the PNS group of neurological score,lower scores were better than those in the untreated group,and the mortality associated with cerebral hemorrhage model group equal. Modeling,48 hours and 7 days,the model group PNS group perihematomal IL-6,integrated optical density of MMP-9 were significantly were higher significantly than those in the sham group(P〈0.05). Modeling 7 days later,the group of PNS integral optical density was lower remarkably than that in the model group(P〈0.05). Conclusion: PNS can reduce neurological deficits and inhibit IL-6,MM-9 release.
