摘要
目的建立反相高效液相色谱法测定氯霉素溶液中氯霉素及其降解产物氯霉素二醇物含量的方法。方法以十八烷基硅烷键合硅胶为填充剂[Dionex Acclaim C18柱(4.6mm×250mm,5μm)];流动相:0.1%庚烷磺酸钠溶液-冰乙酸-乙腈(68:0.2:32);流速:1.0mL/min;检测波长:271nm;柱温:30℃。结果氯霉素与氯霉素二醇物的平均回收率分别为100.6%、100.6%,RSD值分别为1.2%、0.59%m=6);氯霉素的线性范围为25.15-754.48μg/mL(r=0.9998),氯霉素二醇物的线性范围为2.02-60.59μg/mL(r=0.9996)。结论本文所建立的含量测定方法准确可靠、重现性好,可作为制剂的质量控制方法。
Objective To establish a RP-HPLC method for determination of the content of chloramphenicol and glycols in chloramphenicol solution. Methods Dionex Acclaim C18 (5μm; 4.6mm×250mm) was used as the analytical column. Mobile phase: 68 volume of 0.1% W/V sodium 1-heptanesulfonate, 0.2 volume of glacial acetic acid and 32 volume of acetonitrile. The flow rate was 1.0mL/min. The detection wavelength was set at 271nm. The column temperature was 30℃. Results The average recovery and RSD were 100.6% and 1.2% for chloramphenicol(n=6), 100.6% and 0.59% for glycols (n=6). The linear response ranges of chloramphenicol and glycols were 25.15-754.48μg/mL(r=0.9998) and 2.02-60.59μg/mL (r=0.9996). Conclusion The method is good in reproducibility and the result of determination is accurate and reliable. It can be used to determine the content of chloramphenicol and glycols in chloramphenicol solution.
