摘要
目的观察热休克蛋白(hot shock protein,HSP)在早期糖尿病大鼠晶状体上皮细胞的表达及变化,探讨HSP60在糖尿病性白内障病变中的作用。方法鼠龄为6-8周的SD大鼠随机分成糖尿病组及对照组,每组18只,糖尿病组在禁食12 h后一次性腹腔注射10 g·L^-1链脲佐菌素诱发糖尿病模型,成功诱导糖尿病模型后4周、8周、12周分别应用免疫组织化学法及Western blot分析晶状体上皮细胞上HSP60的表达变化。结果对照组大鼠晶状体上皮细胞上未发现有HSP60表达,糖尿病组大鼠在造模后4周、8周、12周均可见晶状体上皮细胞上有HSP60表达,两组间有显著差异(均为P〈0.05)。糖尿病组大鼠造模后4周和8周间表达量无明显差异(P〉0.05);而12周时其表达量开始减少,和4周及8周相比较,差异均有统计学意义(均为P〈0.05)。结论早期糖尿病大鼠晶状体上皮细胞有大量HSP60表达,而且随着时间的延长,表达量呈下降趋势。
Objective To explore the role of hot shock protein 60 (HSP60) in diabetic cataract through observing the expression and change of HSP60 in lens epithelial cells in early diabetic rats. Methods The 6 - 8 weeks old SD rats were divided into normal control group and diabetes group, 18 cases in each group, 10 g·L^-1 STZ were intraperitoneal injected to induce diabetic model in experimental group, At 4 weeks, 8 weeks, 12 weeks after STZ-induced,the expression and change of HSP60 in lens epithelial cells were analyzed by immunohistochemistry and Western blot in normal control rats and diabetic rats. Results There was no HSP60 expression in lens epithelial cells in normal rats. At 4 weeks,8 weeks, 12 weeks after STZ-injected, there were HSP60 expression in lens epithelial cells in diabetic rats, the difference between two groups was statistical significant (all P〈0. 05). The amount of expression began to decrease at 12 weeks after STZ-injected,there were significant differences compared with 4 weeks and 8 weeks ( all P 〈 0.05 ), but there was no significant difference between 4 weeks and 8 weeks (P 〉 0.05 ). Conclusion There are a great number of HSP60 expression in lens epithelial cells in the early diabetic rats, but with the extension of time, the expres- sion quantity decline.
