摘要
目的探讨血管生成素-1(angiopoietin-1,Ang-1)体内抑制视网膜新生血管形成的作用及对血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法高浓度氧诱导C57BL/6J小鼠建立视网膜缺血性病变动物模型。将32只小鼠随机分为4组:正常组、高氧对照组、大剂量治疗组(高氧+100 mg·L-1Ang-1)、小剂量治疗组(高氧+50 mg·L-1Ang-1)。前两组小鼠玻璃体内注射1μL BSS,后两组小鼠玻璃体内分别注射1μL不同浓度的Ang-1。视网膜切片HE染色,计数并比较各组小鼠视网膜新生血管内皮细胞核数。免疫组织化学染色法检测各组小鼠视网膜切片VEGF的表达情况。结果吸入高氧小鼠视网膜均可见突出内界膜的新生血管内皮细胞,发生率为100%。高氧对照组可见突出细胞数为(24.69±2.61)个,大剂量治疗组为(6.56±1.37)个,小剂量治疗组为(10.83±1.81)个。大小剂量治疗组与高氧对照组相比新生血管细胞数明显减少(P<0.01)。在Ang-1治疗的两组中,随着剂量的加大,抑制视网膜新生血管形成的作用逐渐加强。高氧对照组(4.50±0.22)、大剂量治疗组(3.29±0.18)、小剂量治疗组(3.81±0.16)视网膜各层中VEGF表达均较正常组(2.23±0.20)明显增强,差异均有统计学意义(均为P<0.05);与高氧对照组相比,大、小剂量治疗组VEGF表达水平明显降低(P<0.05)。结论 Ang-1能有效抑制高氧诱导的小鼠视网膜新生血管形成,且能减少VEGF的合成和分泌,其抗血管新生作用可能与抑制VEGF表达有关。
Objective To investigate the inhibitory effects of angiopoietin-1 (Ang-1) on retinal neovascularization in vivo and the influence of Ang-1 on vascular endothelial growth factor (VEGF) expression. Methods C57BL/6J mice were put into the environment of high oxygen to establish models of ischemic retinopathy. Thirty- two mice were randomly divided into four groups: normal group, control group, high concentration group ( high oxygen ± 100 mg·L^-1Ang-1 ), low concentration group (high oxygen ±50 mg·L^-1 Ang-1 ). The first two groups received the intravitreal injection of 1 μL PBS,and the other two groups received intravitreal injection of 1 μL Ang-1 with different concentration. HE staining of retina was used to count and compare the endotheliocyte nuclei of new vessels extending from retina to vitreous in the tissue slice. Immunohistochemistry was used to evaluate the expression of VEGF. Results The incidence of retinal neovascular endotheliocyte in the eyes of hyperoxic groups was 100%. The neovascular endotheliocyte in the control group,high and low concentration groups were 24.69 ±2.61,6.56 ± 1.37 and 10. 83 ± 1.81 ,respectively. The number of endotheliocyte nuclei of new vessels in high and low concentration groups were less than those in the control group (P 〈 0.01 ). With the increase of the concentration of Ang-1, the inhibitory effect was enhanced. Compared with the normal group (2.23 ± 0. 20), the expressions of VEGF in the retina were obviously enhanced in the control group (4.50 ± 0,22 ), high concentration group ( 3.29 ± 0. 18 ) and low concentration group ( 3.81 ± 0. 16), there were statistical differences ( all P 〈 0.05 ). Compared with the control group,the expression of VEGF were decreased in the high and low concentration groups (P 〈 0.05 ). Conclusion Ang-1 can inhibit the retinal neovascularization induced by hyperxia, and decrease the expression of VEGF, which may be related with its inhibitory effect on VEGF expression.
