中国汉族人群晚期糖化终产物受体基因-429T/C多态性对其mRNA和蛋白表达的影响研究 被引量:2

Effects of RAGE Gene-429T/C Polymorphism on Its mRNA and Protein Expression among Chinese Han Population

摘要 目的探讨中国汉族人群晚期糖化终产物受体(RAGE)基因-429T/C多态性对其mRNA和蛋白表达的影响。方法选取2012年8—12月糖耐量正常的汉族人群70名,首先通过DNA直接测序分析确定携带TT基因型者57名和携带TC基因型者13名。随机数字表法选取TC基因型组11名,采用匹配原则选择TT基因型组11名,提取外周血单核细胞(PBMC),分别采用实时荧光定量PCR法和Western blotting法检测RAGE mRNA和蛋白的表达水平。结果 TT基因型组RAGE mRNA表达水平高于TC基因型组〔(1.047±0.233)与(0.740±0.209),P<0.05〕。TT基因型组RAGE蛋白表达水平高于TC基因型组〔(1.121±0.252)与(0.916±0.249),P<0.05〕。结论中国汉族人群RAGE基因-429T/C多态性可能影响RAGE mRNA和蛋白的表达,-429C等位基因可能降低RAGE基因表达水平。 Objective To investigate the effects of RAGE gene- 429T/C polymorphism on its mRNA and protein expression in a Chinese Han population. Methods A total of 70 Han people with normal glucose tolerance( NGT) were selected as study subjects from August 2012 to December 2012,by DNA sequencing analysis,TT genotype was found in 57 subjects,and TC genotype was found in 13 subjects,11 subjects with TT genotype and 11 subjects with TC genotype were selected. The mRNA and protein expression of RAGE in PBMC were evaluated by real- time PCR and western- blot analysis,respectively. Results The expression levels of RAGE mRNA in the subjects with TT genotype were significantly higher than those in the subjects with TC genotype 〔( 1. 047 ± 0. 233) vs.( 0. 740 ± 0. 109),P〈0. 05〕. The expression levels of RAGE protein in the subjects with TT genotype were also significantly higher than those in the subjects with TC genotype 〔( 1. 121 ± 0. 252) vs.( 0. 916 ± 0. 249),P〈0. 05 〕. Conclusion The RAGE gene- 429 T / C polymorphism may influence expression of RAGE mRNA and protein,- 429 C allele may reduce RAGE gene expression levels.
