摘要
研究了以茶蛋白为原料制备高F值低聚肽过程中的酶解工艺,采用两步定向酶切的方式,探讨合适的蛋白酶种类,并通过单因素试验考察相关工艺条件对酶切效果的影响。结果表明:碱性蛋白酶和复合风味蛋白酶联用对茶蛋白的水解效果较好;一级定向酶切的最适工艺条件为:底物浓度5%、加酶量(E/S)4%、反应液pH 9.0、反应温度50℃、反应时间3h,此条件下茶蛋白的水解度达到36.02%;二级定向酶切的最适工艺条件为:加酶量(w/v)0.2%、反应液pH值6.5、反应温度55℃、反应时间2h,此条件下水解液的OD220/OD260值达到2.53。该试验为后续水解液分离纯化制备高F值低聚肽产品奠定了基础。
In order to study the enzymatic hydrolysis process during high fischer ratio oligopeptides preparation with tea protein as raw material,a two-step method for selective enzymatic hydrolysis was used and the types of proteases and technical conditions in each step were discussed.Results show that the hydrolysis effect of tea protein is good by using alcalase and flavourzyme for the two-step method.The optimum conditions of the first-order selective enzymatic hydrolysis are substrate concentration5%,enzyme dosage(E/S)4%,pH9.0,reaction temperature 50 ℃,reaction time 3 h.Under these conditions,the degree of hydrolysis of tea protein is up to 36.02%.The optimum conditions of the second-order selective enzymatic hydrolysis are enzyme dosage(w/v)0.2%,pH6.5,reaction temperature 55 ℃,reaction time 2 h.Under these conditions,the value of OD220/OD260 reaches 2.53.The results of this paper lay a foundation for the further study on hydrolysate purification and high fischer ratio oligo-peptides products producing.
