蛭龙活血通瘀胶囊对脑出血大鼠HIF-1α与Bcl-2蛋白表达的影响 被引量:2

摘要 目的:在不同时间点观察大鼠脑出血后Bcl-2和HIF-1α蛋白的表达情况及经蛭龙活血通瘀胶囊干预对其的影响,为蛭龙活血通瘀胶囊治疗脑出血提供参考。方法:将100只SD大鼠随机分为蛭龙活血通瘀胶囊高剂量组、蛭龙活血通瘀胶囊中剂量组、蛭龙活血通瘀胶囊低剂量组、模型组和假手术组各20只。采用自体血注入法制备脑出血大鼠模型,根据脑出血术后不同时间点,每组再继续分为术后6 h、1 d、3 d、7d 4个亚组,每亚组5只。术前1周开始灌胃,每天1次。蛭龙活血通瘀胶囊高剂量组、蛭龙活血通瘀胶囊中剂量组、蛭龙活血通瘀胶囊低剂量组予以等体积相应浓度的蛭龙活血通瘀胶囊灌胃,给药剂量分别为1.6 g·(kg·d)-1、0.8 g·(kg·d)-1、0.4 g·(kg·d)-1;模型组和假手术组均给予等体积的生理盐水灌胃。免疫组化法测定HIF-1α与Bcl-2蛋白的表达。结果:在相应时间点,蛭龙活血通瘀胶囊中剂量组、蛭龙活血通瘀胶囊高剂量组与蛭龙活血通瘀胶囊低剂量组比较,其HIF-1α与Bcl-2蛋白的表达均升高,差异有统计学意义(P<0.05);在相应时间点与模型组比较,蛭龙活血高剂量组、蛭龙活血中剂量组、蛭龙活血低剂量组Bcl-2与HIF-1α蛋白的表达升高,差异有统计学意义(P<0.05)。结论:蛭龙活血通瘀胶囊能明显促进HIF-1α与Bcl-2表达,HIF-1α与Bcl-2蛋白之间存在正相关性。在脑出血大鼠中,蛭龙活血通瘀胶囊可能有一定的脑保护作用,且存在量效关系。 Objective: To observe dynamically the intervention effect of Zhilong Huoxue Tongyu Capsule( ZLHXTYJN) on the protein expression of HIF-1α and Bcl-2 in intracerebral hemorrhage rats and to explore the new proprietary Chinese medicine brain protective agent to treat the disease to provide the reference. Methods: Using SD rats as the research object,they were randomly divided into five groups: high dose ZLHXTYJN group,moderate dose ZLHXTYJN group,low dose ZLHXTYJN group,model group and sham operation group. Autologous blood injection method was used to produce cerebral hemorrhage model. Using immunohistochemical method to determine the expression of HIF-1α and Bcl-2 protein. Results: Compared with the low dose ZLHXTYJN group,the expression of HIF-1α and Bcl-2 protein of moderate dose ZLHXTYJN group and high dose ZLHXTYJN group significantly raised( P〈0. 01); compared with the model group,the expression of HIF-1α and Bcl-2 protein of each ZLHXTYJN group significantly raised( P〈0. 01). Conclusion: ZLHXTYJN can obviously promote HIF-1α and Bcl-2 expression. There is a positive correlation between HIF-1 α and Bcl-2 protein. ZLHXTYJN in rats may be mediated by HIF-1 α role to regulate the expression of Bcl-2 and inhibit the apopt-osis of cerebral hemorrhage,which has cerebral protective effects on cerebral he-morrhage in rats,and there exists a certain dose-effect relationship.
