芍药汤对胃肠湿热型溃疡性结肠炎大鼠结肠黏膜核因子-κB DNA结合活性的影响 被引量:16

摘要 目的研究芍药汤对胃肠湿热型溃疡性结肠炎大鼠核因子-κB(NF-κB)DNA结合活性的影响。方法将40只SD大鼠随机分为4组:正常组,模型组,芍药汤组,柳氮磺胺吡啶(SASP)组。制备胃肠湿热型模型,然后采用三硝基苯磺酸(TNBS)制备灌肠法溃疡性结肠炎模型,灌胃治疗10 d后处死大鼠,取新鲜结肠黏膜标本提取核蛋白,利用以酶联免疫吸附测定(ELISA)为基础的Trans AMTMNF-κB P65试剂盒检测各组NF-κB相对活性。结果与正常组比较[(0.263±0.041)],模型组大鼠NF-κB相对活性明显增高[(0.467±0.083)],差异有高度统计学意义(P<0.01);与模型组比较,芍药汤组大鼠结肠黏膜NF-κB相对活性明显降低[(0.271±0.089)],差异有高度统计学意义(P<0.01)。结论芍药汤能明显降低胃肠湿热型溃疡性结肠炎大鼠模型结肠黏膜NF-κB相对活性,可能是其治溃疡性结肠炎作用的机制之一。 Objective To observe the effect of Peony Decoction Decoction on DNA binding activity of NF-κB in colonic mucosa of gastrointestinal syndrome with ulcerative colltis rats. Methods 40 rats were randomly divided into four groups, normal group, model group, Peony Decoction group, SASP group. Gastrointestinal syndrome rat model was prepared, then ulcerative colltis rat model was made by trinitrobenzene-sulfonic acid(TNBS). The rats were killed to get fresh colonic mucosa and extract nuclear proteins after 10 days of treatment. Relative activity of NF-κB was detected by ELISA with a Trans AMTMNF-κB p65 kit. Results Compared with normal group [(0.263±0.041)], the relative activity of NF-κB was significantly higher in model group [(0.467±0.083)], the difference was statistically significant(P 〈0.01); compared with model group, the relative activity of NF-κB was significantly lower in Peony Decoction group [(0.271±0.089)],the difference was statistically significant(P 〈0.01). Conclusion Peony Decoction can inhibit the relative activity of NF-κB in colonic mucosa of gastrointestinal syndrome with ulcerative colltis rats. The inhibitory effect of Peony Decoction on ulcerative colltis maybe associate with the inhibition of NF-κB activity.
