摘要
目的观察烟酰胺超载与氧化应激之间的关系,以确定烟酰胺在机体氧化性损伤中扮演的重要角色。方法利用长期大鼠高烟酰胺喂养模型,结合Real time PCR和Western blot方法,研究烟酰胺超载对氧化应激相关分子水平的影响。结果在烟酰胺的作用下,NNMT、DNMT1、PARP1、Catalase及P53的表达水平呈现剂量依赖性的提高;同型半胱氨酸代谢酶BHMT、CBS与MTR出现了显著的表达补偿机制;INS1的表达受烟酰胺的影响较小。结论烟酰胺超载会触发氧化应激,这种改变可能全面并直接参与代谢综合征及其临床结局的形成和发展。
Objective The aim of this study was to investigate the effect ofnicotinamide on the oxidative stress injury in rats. Methods Long-term high nicotinamide feeding test, together with real time PCR and Western blot were performed to investigate the effect ofnicotinamide on oxidative stress injury in rats. Results Nicotinamide loading induced a significant increase in the levels of NNMT, DNMT1, PARP1, Catalase and P53; There is a compensation mechanism for homocysteine metabolism enzyme (BHMT, CBS and MTR); However, INS1 was less affected by nicotinamide. Conclusion Excess nicotinamide may trigger oxidative stress which may be directly or indirectly involved in the development of metabolic syndrome and its clinical outcomes.
