液质联用法测定抗结核药物血药浓度 被引量:3

The detection of antituberculosis drugs concentration in plasma by HPLC-MS/MS

摘要 目的建立利用高效液相色谱质谱联用技术(HPLC-MS/MS)同时测定血浆中异烟肼(H)、利福平(R)、吡嗪酰胺(Z)、乙胺丁醇(E)4种药物浓度的方法。方法采用Waters Xterra C18色谱柱(3.5μm 2.1×150mm);以0.01mol/L的乙酸铵:乙腈:甲醇(35∶10∶55)为流动相;流速0.2ml/min;柱温:室温;进样量为10μl。使用电喷雾离子源(ESI),多反应离子监测(MRM)。结果 HRZE的线性范围分别为4.004-200.2ng/ml、20.06-1003ng/ml、20.12-1006ng/ml、0.8012-40.06ng/ml,检测限分别为1ng/ml、2ng/ml、2ng/ml、0.2ng/ml,RSD分别为1.74%、1.68%、1.46%、1.32%。结论该方法同时测定血浆中HRZE的血药浓度线性关系良好,结果稳定可靠。 Objective To establish a method for detecting the concentration of RFP,PZA,INH and EMB in plasma by HPLC-MS/MS simultaneously. Methods Column:Waters Xterra C18 (3.5μm 2.1×150mm) ;Mobile phase:0.01mol/L ammonium acetate : acetonitrile : methanol ( 35:10:55 ) ; Flow rate : 0.2ml/min ; column temperature : room temperature ; the injec- tion volume was lOlL. Electrospray ionization (ESI) , multiple reaction monitoring (MRM). Results The linear ranges of HRZE were 4.004 - 200.2ng/ml,20.06 - 1003ng/ml,20. 12 - 1006ng/ml,0. 8012 - 40.06ng/ml, the detection limit were l ng/ml,2ng/ml,2ng/ml,0.2ng/ml, and RSD were 1.74%, 1.68%, 1.46%, 1.32%. Conclusion The method could be used to determine the content of HRZE in human plasma.
