自噬途径在人支气管上皮细胞恶性转化中的作用机制

摘要 目的对自噬途径在人支气管上皮细胞恶性转化中的作用机制进行分析与探讨。方法利用激光共聚焦扫描显微镜成像和FRET技术,在活细胞中实时研究结晶型Nis与反式-BPDE恶性转化16HBE细胞中自噬途径的作用机制。结果正常细胞与恶性转化细胞相比,GEP-LC3蛋白的点状聚集明显减少,仅仅达到16HBE-N细胞的1/3,由此充分表明,蛋白质印迹法检测发现,自噬水平下降,m TOR激酶活性上升,Beclin1表达降低。结论研究表明,反式-BPDE会通过自噬途径参与诱导16HBE细胞恶性转化的癌变过程中,为肺癌的预防与治疗提供重要信息。
