摘要
目的优化金樱子中鞣质的超声提取工艺。方法采用磷钼钨酸-紫外分光光度法测定鞣质含量,在单因素试验的基础上,选取液料比、超声提取时间和乙醇百分浓度作为自变量,金樱子鞣质提取率为因变量,对自变量各水平进行多元线性回归和二项式拟合,采用星点设计-响应面法选择最佳工艺条件,并进行预测分析。结果最佳工艺为:乙醇浓度75%,料液比1∶30,超声提取时间60 min,提取温度为50℃。结论优化的金樱子中鞣质超声波提取工艺简便可靠、稳定、提取率高,为金樱子中鞣质的开发利用奠定基础。
Objective To optimize the process of ultrasonic extracting effective constituents from Rosa Laevigatae Fruc- tus of tannin. Methods The content of tannin was determined by phosphomolybdotungsitc acid-UV spectmphotometry sol- vent. Based on single-factor tests, with ethanol percent, solid-liquid ratio, extraction time as independent variables and yield of tannin from Rosa Laevigatae Fructus as dependent variable, multiple linear regression and binomial fitting was adopted by central composite design model, and determine optimum extraction technology, and the forcast analysis was done. Results The best extraction process was as followed: the concentration ethanol of 75%, ratio of solid-liquid of 1: 30, extraction time of 60 min, extraction temperature of 50℃. Conclusion This optimized extraction process is simple and reliable, it can laid a certain foundation for development and utilization of tannins from Rosa Laevigatae Fructus.
