摘要
目的测定不同采收期唇香草中5种化学成分的动态变化,初步确定唇香草的最佳采收期。方法采用20%二甲基亚砜甲醇溶液提取唇香草,利用高效液相色谱法,以0.3%甲酸甲醇-0.3%甲酸水为流动相进行梯度洗脱,以290 nm为检测波长,同时测定不同生长期唇香草中咖啡酸、迷迭香酸、地奥司明、蒙花苷以及胡薄荷酮5种化学成分的含量。结果在上述色谱条件下,5个化学成分的线性关系、分离度、重复性以及回收率等均符合要求,不同采收期的唇香草中均是蒙花苷和地奥司明的含量较高,胡薄荷酮的含量居中,咖啡酸和迷迭香酸含量较低;5种化学成分在唇香草不同采收时期的含量存在差异,但在开花前期和开花期5种化学成分的含量均较高。结论可初步确定唇香草的最佳采收期为开花期,其化学成分的动态变化规律可为唇香草的规范化种值(GAP)生产提供基础数据。
Objective To measure the dynamic changes of five chemical components of Ziziphora clinopodioides Lam. at different growth stages, and to determine the optimal harvesting time.Methods The Ziziphora clinopodioides Lam. was extracted from 20 % dimethyl sulfoxide-methanol solution. The method of High Performance Liquid Chromatography( HPLC) was used,with the 0. 3%mobile phase of formic acid-methanol-0. 3% formic acid on a gradient elution,to detect the Caffeic acid, Rosemary acid, Diosmin, Linarin and Pulegone in different growth periods of Ziziphora clinopodioides Lam. at 290 nm wave length at the same time. Results With these chromatographic parameters,the linear relationship,degree of separation,precision,stability,repeatability and recovery rate of the five components were all qualified. The contents of Linarin and Diosmin were the highest,with Pulegone second to it while the Caffeic acid and Rosemary acid's contents were lower at different growth periods. And these five chemical components in Ziziphora clinopodioides Lam. varied at different growth periods. But all of them were higher in pre-flowering period and flowering period. Conclusion According to the data, we can determine the flowering period was the optimal harvesting time for Ziziphora clinopodioides Lam. initially. The pattern of dynamic changes of the chemical components of Ziziphoraclinopodioides Lam. could provide the basic data for the Good Agriculture Practice( GAP) of Ziziphora clinopodioides Lam.
