摘要
目的:探讨脑5-羟色胺(5-HT)及其旁路关键酶在术后疲劳综合征(POFS)大鼠模型中的变化。方法:将64只SD实验大鼠随机分为模型组和对照组(各分为术后1、3、7、10 d组),模型组行70%中段小肠切除端端吻合术,对照组仅行开关腹手术。分别于术后1、3、7、10 d取脑组织,采用高效液相色谱-串联质谱法测定海马组织中5-HT的含量,荧光定量PCR法检测海马组织中色氨酸羟化酶1、2(TPH1、TPH2)和吲哚胺-2,3-二加氧酶(IDO)的表达量。结果:与对照组相比,模型组大鼠5-HT的含量分别于术后1、3 d明显升高(P<0.05);在模型组大鼠中,术后1、3 d 5-HT的含量均较术后7、10 d明显升高(P<0.05),术后3 d升至最高(P<0.05),术后7、10 d逐渐恢复正常。与对照组相比,模型组大鼠TPH1的表达量于术后3 d明显升高(P<0.05);在模型组大鼠中,术后3 d TPH1的表达量较术后1、7、10 d明显升高(P<0.05)。与对照组相比,模型组大鼠TPH2的表达量分别于术后1、3 d明显升高(P<0.05);在模型组大鼠中,术后1、3 d TPH2的表达量均较术后7、10 d明显升高(P<0.05),术后3 d升至最高,术后7、10 d逐渐恢复正常。与对照组相比,模型组IDO的表达量于术后1 d明显升高(P<0.05);在模型组大鼠中,术后1 d IDO的表达量较术后3、7、10 d明显升高(P<0.05)。结论:在POFS大鼠模型中,5-HT的含量术后早期明显升高,其升高与5-HT限速酶TPH1、TPH2的表达量上调密切相关。
Objective:To explore the role of changes of key enzymes of brain 5-HT and 5-HT bypass in rat model of postoperative fatigue syndrome. Methods:64 SD rats were randomly divided into model group and con-trol group according to the weigh (which both including 1 d, 3 d, 7 d, 10 d group). The rats of model group would be resected the middle 70%small bowel, and control group only undergoing on-off abdominal surgery. The brains were removed on 1 d, 3 d, 7 d, 10 d postoperatively respectively, and we detected the concentration of 5-HT by high performance liquid chromatography and the mRNA expression of TPH1, TPH2, IDO by realtime lfuores-cence quantitative PCR in hippocampus area. Results:Compared with control group, concentration of 5-HT of model group rats increased signiifcantly on 1 d, 3 d postoperatively respectively (P〈0.05). And in model group rats, concentration of 5-HT on 1 d, 3 d increased signiifcantly respectively compared with 7 d, 10 d postoperative-ly (P〈0.05), on 3 d to its highest (P〈0.05), 7 d, 10 d gradually returned to normal. Compared with control group, mRNA expression of TPH1 in model group rats increased signiifcantly on 3 d postoperatively (P〈0.05). And in&amp;nbsp;model group rats, mRNA expression of TPH1 on 3 d increased signiifcantly compared with 1 d, 7 d, 10 d post-operatively (P〈0.05). Compared with control group, mRNA expression of TPH2 in model group rats increased signiifcantly on 1 d, 3 d postoperatively respectively (P〈0.05). And in model group rats, mRNA expression of TPH2 on 1 d, 3 d increased signiifcantly respectively compared with 7 d, 10 d postoperatively (P〈0.05), on 3 d to its highest, 7 d, 10 d gradually returned to normal. Compared with control group, mRNA expression of IDO in model group rats increased signiifcantly on 1d postoperatively (P〈0.05). And in model group rats, mRNA expres-sion of IDO on 1 d increased signiifcantly compared with 3 d, 7 d, 10 d postoperatively (P〈0.05). Conclusion:In rat model of postoperative fatigue syndrome, concentration of 5-HT increases signiifcantly early postopera-tively. Its mechanism refers to up-regulation of mRNA expression of TPH1, TPH2 of 5-HT rate-limiting enzyme.
