摘要
目的建立HPLC同时测定野葛、粉葛、泰国葛中异黄酮葛根素、大豆苷和染料木素含量的方法。方法色谱柱为Hypersil BDS C18柱(250 mm×4.6 mm,5μm);流动相为甲醇-水,梯度洗脱,流速0.7 m L/min,进样量10μL,柱温25℃,检测波长250 nm。结果葛根素、大豆苷和染料木素分别在0.05~1.20μg,0.025~0.60μg,0.025~0.60μg范围内呈良好线性关系,r分别为0.9999,0.9999,0.9999。其平均回收率为97.26%~104.64%,RSD均小于3%。结论建立的方法简单、快捷,重复性好,结果准确可靠,可为葛根的资源研究提供支持。
Objective To establish a HPLC method for simultaneous determination of puerarin, daidzin, and genistein from Pueraria lobata (wild) Ohwi, Pueraria thomsonii Benth and Pueraria mirifica. Methods The Hypersil BDS C18 column (250 mm×4.6 mm, 5 μm) was adopted. The mixture of methanol and water was used as mobile phase of gradient elution, with the flow rate of 0.7 mL/min. The injection volume was 10 μL. The Column temperature was 25 ℃, and the detection wavelength was 250 mn. Results This method had good linearity in the range of 0.05-1.20 μg, 0.025-0.60 μg and 0.025-0.60 μg for puerarin, daidzin and genistein, respectively, with r of 0.9999, 0.9999 and 0.9999, respectively. The average recoveries were 97.26 %-104.64 %, and RSDs were less than 3 %. Conclusion The method is simple, fast, reproducible, and with accurate and reliable results, which can provide a reference for the study of Radix puerariae resource.
