低压缺氧环境对大鼠胃肠运动影响机制及生白术的干预作用 被引量:8

Effects and mechanism of raw atractylodes on gastrointestinal movement of rats in the low pressure and hypoxia environment

摘要 [目的]观察低压缺氧Wistar大鼠的胃窦组织、Cajal间质细胞及小肠推进率,探讨低压缺氧胃肠运动功能障碍发生机制和生白术的干预作用。[方法]70只Wistar大鼠随机分为7组,对照组(兰州海拔,1 430m,A组),模型组(3 500m海拔,B1组;5 000m海拔,B2组)、马来酸曲美布汀干预组(3 500m海拔,C1组;5 000m海拔,C2组)、生白术干预组(3 500m海拔,D1组;5 000m海拔,D2组)。每组10只,除对照组,其余6组依据不同海拔高度利用低压氧舱造模3d,其中C、D组每日分别给予马来酸曲美布汀和生白术灌胃。3d后各组大鼠均给予2ml墨汁灌胃,30min后处死,测算墨汁的小肠推进率,观察胃窦组织病理变化,免疫组织化学方法检测胃窦组织Cajal间质细胞(ICC)的阳性面积。[结果]与A组比较,B1组小肠推进率及胃窦ICC阳性面积减少,组织黏膜表层见坏死和脱落、腺体组织减少伴水肿、炎性细胞浸润;B2组小肠推进率及ICC阳性面积增加,黏膜组织坏死脱落加重、腺体组织减少和水肿加重,有大量炎性细胞浸润(P<0.05)。C1组ICC阳性面积及小肠推进率升高,但P>0.05,C2组ICC阳性面积及小肠推进率下降(P<0.05),C1和C2组织黏膜炎性细胞浸润明显减轻,但组织黏膜坏死和脱落变化不明显。D1组ICC阳性面积和小肠推进率升高,D2组ICC阳性面积和小肠推进率减少,D1和D2组织黏膜坏死脱落和炎性细胞浸润都明显减轻(P<0.05)。[结论]低压缺氧导致胃肠道运动功能紊乱,与海拔高度关系密切,与高原环境对胃窦组织黏膜损伤及ICC阳性面积变化有关,生白术可有效干预胃肠运动功能紊乱,修复损伤的黏膜组织。 [Objective]To evaluate the interstitial cells of Cajal and small intestine propulsion rate in the rats under low pressure and hypoxia environment,investigate the mechanism of the gastrointestinal motility dysfunction and the effects of raw atractylodes.[Methods]Seventy Wistar rats were randomly divided into 7groups:control group(1 430 mabove sea level,A group),model group,Maleic acid trimebutine group,raw atractylodes group at 3 500 mand 5 000 mabove sea level(B1and B2,C1 and C2,D1 and D2 group).They were managed for 72 hin different altitude levels by hypobaric oxygen chamber,and lavaged with 2ml ink after 72 h,and put to death 30 min after ink lavage.Small intestinal propulsion rates of ink were measured.Pathological changes of gastric antrum tissue and the positive area rates of interstitial cells of Cajal were observed.[Results]Compared with group A,the positive area rates of ICC reduced and small in-testinal propulsion rates downregulated in group B1,C2 and D2,while those were increased in group B2,C1 and D1.But those of group C1 and C2 were not significantly improved compared with group B1 and B2.Those of group D1 and D2were significantly improved compared with group B1 and B2.There were necrosis,shedding,edema and inflammatory cells infiltration of mucosal surface in organization in group B1 and B2.They were relieved in C and D,but were not significant in group C.[Conclusion]The low pressure and hypoxia environment could induce gastrointestinal motor function disorder,which were closely associated with the altitude and the positive area rates of ICC.Raw atractylodes could effectively adjust the disorder and repair injury.
