超声波辅助提取分离菠菜色素的工艺研究 被引量:7

摘要 利用超声波辅助法提取菠菜中的色素,通过单因素实验和正交试验探索了提取剂、料液比、提取温度、超声波时间和超声波功率对提取效果的影响,得到了提取菠菜色素工艺的最优条件。实验结果表明,以乙醇与丙酮混合液(体积比为1∶1)为提取剂,料液比为1∶5(g·m L-1),提取温度为50℃,超声波时间为30 min,超声波功率为280 W,菠菜色素的产率为0.187 mg·g-1。根据胡萝卜素、叶黄素、叶绿素极性不同,用层析柱法分离菠菜色素,既能达到很好的分离效果,又能循环利用溶剂。 Pigment was extracted from the spinach leaves by the of ultrasonic-assisted method,and the effects of extracting reagent,the ratio of material to fluid,extraction temperature,ultrasonic time and power were studied by the single factor experiment and orthogonal test. The result showed that the best extraction conditions of spinach pigment was follows:ethanol and acetone(volume ratio of 1 ∶1) as the extracting reagent,the ratio of material to fluid 1 ∶5(g·m L^-1),extraction temperature 50 ℃,ultrasonic power280 W and ultrasonic time 30 min,and the yield of spinach pigment was up to 0.187mg·g^-1. According to the different polarities of the carotene,lutein and chlorophyll,spinach pigment was separated by chromatography column,which could achieve a good separation effect and the solvent was recycled.
