摘要
TCF4属核内转录因子,在Wnt/β-catenin信号途径中可调控下游基因表达。TCF4有着分子(开关)的双重作用,可表现为转录激活或转录抑制的作用,影响一系列基因的表达,在胚胎的发育及肿瘤的发生发展中起重要的作用。TCF4在转录后加工过程中存在选择性剪接,可形成多种mRNA剪接异构体,编码产生不同功能结构域的蛋白,进而对下游多种基因的转录进行活性调节。TCF4及其剪接异构体的异常调控与肿瘤的发生发展密切相关。
TCF4 is a nuclear transcription factor,which can regulate the expression of downstream genes in the Wnt / β-catenin signaling pathway.As a molecular switch,TCF4 can activate or repress gene expression depending on its binding proteins,which influences a series of genes expression involved in embryo development and tumorigenesis.TCF4 mRNA is alternative spliced during its maturation,which generates various splicing isoforms encoding proteins with different functional domains,thus to regulate the activity of transcription of multiple downstream genes.The abnormal regulation of TCF-4 and its splicing isoforms have closely relationships with the tumors.
