摘要
目的:探讨在2型糖尿病的发生、发展的不同阶段施加运动干预,对大鼠骨代谢指标及骨髓瘦素和瘦素受体蛋白表达的影响,为运动预防和治疗2型糖尿病骨代谢异常提供理论依据。方法:刚断乳雄性SD大鼠随机分为5组,正常对照组(NCG),糖尿病对照组(DCG),治疗运动组(ZLG),预防运动组(YFG),防治运动组(FZG),每组各11只大鼠。除NCG组使用普通饲料喂养外,其余各组全程使用高脂高糖饲料喂养。YFG组和FZG组在2型糖尿病造模前进行为期6周的游泳训练;造模后对ZLG组、FZG组进行为期6周的游泳训练。各组于运动结束后24 h内,禁食12 h后取样,检测糖、骨代谢相关指标,同时检测各组骨髓瘦素和瘦素受体蛋白表达。结果:1)体重变化方面,ZLG组、YFG组和FZG组与DCG组比,体重均有下降的趋势,其中ZLG组和FZG组有统计学意义(P<0.05)。2)糖代谢方面,与DCG组相比,ZLG组胰岛素显著性下降(P<0.05),FZG组血糖、糖化血清蛋白和胰岛素值均有显著性下降(P<0.01,P<0.05,P<0.01)。3)骨代谢方面,与DCG组相比,FZG组抗酒石酸酸性磷酸酶值显著下降(P<0.01);FZG组骨钙素值显著升高(P<0.01)。4)骨髓瘦素、瘦素受体蛋白表达方面,与DCG组相比,ZLG组、YFG组及FZG组骨髓瘦素含量有下降趋势,其中只有FZG组变化有统计学意义(P<0.01);ZLG组、YFG组及FZG组骨髓瘦素受体含量有升高趋势,其中只有FZG组变化有统计学意义(P<0.01)。结论:在糖尿病发病不同阶段施加运动干预,可以不同程度改善2型糖尿病大鼠糖代谢紊乱,促进大鼠骨代谢调整,缓解瘦素抵抗。在糖尿病发病全程施加运动干预效果最佳。
Purposes:To provide the theoretic evidence for prevention and treatment for type 2 diabetes by discussing how does interference of physical exercise influence rats with type 2 diabetes and its marrow leptin and leptin receptor protein expression in different stage of morbidity. Method:To randomly divide the male SD rats (Weaned) into five groups, each group compromises 11 rats. For instance, group of normal comparison( NCG), group of diabetes comparison( DCG), group of physical exercise treatment (ZLG), group of physical exercise prevention (YFG), group of FZG. Except rats from NCG group fed on normal stuff,the rest groups are fed on fatty and sugary stuff and inject a small does of STX on sixth week, resulted the type 2 diabetes. Group YFG and group FZG are trained to swing for six weeks before the morbidity of type 2 diabetes ;After the morbidity of type 2 diabetes, group ZLG and group FZG are trained to swing for six weeks. After all groups finished the physical exercises ,they are sampled after 12 hours of jejunitas within 24 hours such as FPG, glycocylat- ed serum protein ( GSP ), Insulin, Osteocalcin (OS) and tartrate resistant acid phosphatase (TRACP) as well as expres- sion of bone metabolism and receptor protein. Results : 1. As for weight, the group ZLG, YFG decreased in comparison with group FZG and group DCG, among which the group ZLG and group FZG have statistical significance (P 〈 0.05 ). 2. As for glycometabolism,FPG of group ZLG obviously decreased ( P 〈 0.05 ) compared with group DCG. Group YFG' s FPG, GSP and Insulin slightly decreased while group FZG decreased sharply in terms of what mentioned above( P 〈 0.01, P 〈 0. 05, P 〈 0.01 ). 3. In terms of bone metabolism, group ZLG, group YFG and group FZG' s tartrate resistant acid phosphatase (TRACP) presented a decrease, among which the group FZG has statistical significance ( P 〈 0.01 ) ; group ZLG, group
