钼蓝分光比色法测定聚合人脐带血红蛋白溶液中还原性维生素C的含量 被引量:2

Determination of reduced vitamin C content in polymerized hemoglobin solutions in humen pleconte by Molybdenum blue spectrophotometry

摘要 目的建立1种适用于聚合人脐带血红蛋白(HBOCs)体系的还原性维生素C(Vc)的检测方法。方法采用钼蓝分光比色法对HBOCs样品中的还原性Vc含量进行检测,研究盐酸对HBOCs的沉淀效果.在测定过程中摸索最大吸收波长、盐酸加量和反应时间的优化条件,并在此基础上对钼蓝分光比色法的精密度、重复性和回收率作了考察。结果当酸沉液中盐酸浓度>1mol/L时对HBOCs样品有着良好的沉淀效果;显色反应条件确定为1-2mL盐酸加量,反应时间>60min,最大吸收波长值710nm。在此条件下,Vc加量在0-200μg的标准曲线线性度达0.9985;精密度和重复性试验结果良好,RSD值均<5%;HBOCs中Vc回收率为97%-102%。结论钼蓝分光比色法可以有效测定HBOCs溶液中的Vc含量,具有操作简单、成本低等特点。 Objective To build a testing method for estimating the reduced vitamin C(Vc) content in polymerized hemoglobin-based oxygen carriers solutions(HBOCs) human placenta.Methods Molybdenum blue spectrophotometry method was used to determine the reduced amount of Vc contents in HBOCs samples.After precipitation of HBOCs by the addition of hydrochloric acid(HCl),the following effects of the color of the reaction in solution were optimized: maximum absorption wavelength,volume of HCl addition and reaction time.The experiments on precision and repeatability and Vc recovery were then conducted.Results HBOCs could be fully precipitated by acidic solutions with HCl concentrations above 1.0mol/L.The color of the reaction was optimized when 1- 2mL HCl were added for 60 minutes of reaction time at a maximum absorption wavelength of 710 nm.The standard curve was confirmed in the range of addition of 0- 200μg Vc with a linear correlation coefficient,R = 0.998 5.The relative standard deviation(RSD) of both precision and repeatability experiments were below 5%.The coefficients of Vc recovery in HBOCs samples were in the range of 97%- 102%.Conclusion Reduced Vc contents could be tested effectively by molybdenum blue spectrophotometry method,which could be used in the development of HBOCs due to simple operation and low cost.
