潜在致病性的弗氏枸橼酸杆菌的筛查 被引量:1

摘要 目的筛查具有潜在致病性的弗氏枸橼酸杆菌。方法对从河南省睢县分离到的36株弗氏枸橼酸杆菌进行黏附HEp-2细胞的检测,以及与HEp-2细胞(MOI:100)作用10 h后的乳酸脱氢酶(LDH)释放情况分析。同时比较弗氏枸橼酸杆菌CF74(Citrobacter freundii 74)、CF72(Citrobacter freundii 72)、肠聚集性大肠埃希菌(Enteroaggregative E.coli,EAEC)、肠致病性大肠埃希菌(Enteropathogenic E.coli,EPEC)2348/69和HB101的黏附和细胞毒性差异。结果 16株弗氏枸橼酸杆菌几乎没有黏附性,黏附指数<1;18株弗氏枸橼酸杆菌具有中等强度的黏附性,黏附指数<100;2株弗氏枸橼酸杆菌CF74和CF65(Citrobacter freundii 65)具有强的黏附性。与HEp-2细胞作用10 h后,除了CF74(24.4%),其余的35株弗氏枸橼酸杆菌具有低的LDH释放量,LDH释放量与阴性对照HB101(5.02%)相似。说明除了CF74,其余的弗氏枸橼酸杆菌不具有细胞毒性。CF74具有和EAEC042一样的聚集性黏附类型,并且CF74引起的LDH释放率明显高于CF72、2348/69和HB101(P<0.01)而低于EAEC17-2。结论作为潜在的致病菌,CF74具有强的黏附性和细胞毒性。 Objective To screen potential pathogenic Citrobacter freundii. Methods The adhesion of C. freundii on HEp-2 cells w as tested by using 36 C. freundii strains isolated from Suixian county,Henan province,and lactate dehydrogenase( LDH) released induced by C. freundii w as detected after co-cultured w ith HEp-2 cells for 10 hours.M oreover,the adhesion and cytotoxicity w ere compared among C. freundii 74,C. freundii 72,enteroaggregative E. coli( EAEC) strain 042 and 17-2,enteropathogenic E. coli( EPEC) strain 2348 /69 and E. coli strain HB101.Results Sixteen C. freundii strains show ed little adhesion, w ith indexes 〈 1; 18 C. freundii strains show ed intermediate adhesion,w ith adhesion indexes ranged from 1 to 100; C. freundii 65 and C. freundii 74 show ed the strongest adhesion,w ith adhesion indexes 〉 100. After 10 hours co-culturing w ith HEp-2 cells,35 C. freundii strains,except C. freundii 74( 24%), show ed low LDH releasing, similar to that induced by positive control HB101(5. 02%). It suggested that all the isolates,except C. freundii 74 were likely to be non-cytotoxic. C. freundii 74 show ed an aggregative adherence pattern similar to that of EAEC 042,the LDH releasing induced by C. freundii 74 w as higher than C. freundii 72,2348 /69 and HB101( P 〈 0. 01),but low er than EAEC17-2. Conclusion As a potential pathogen,C. freundii 74 show ed the strongest adhesion and cytotoxicity.
