摘要
目的:建立陕西产黄花油点草中烟花苷含量测定的方法。方法:采用高效液相色谱法。色谱柱为Thermo Hypersil GOLD,流动相为乙腈-0.4%磷酸水溶液(梯度洗脱),流速为1 ml/min,柱温为20℃,检测波长为360 nm。结果:烟花苷的进样量在5.9~35.3μg范围内与其峰面积呈良好线性关系(r=0.999 5);精密度、重复性、稳定性试验的RSD〈2%;平均加样回收率为98.9%,RSD为1.6%(n=6)。结论:该方法稳定、可行,可用于陕西产黄花油点草水提取物及总黄酮部位中烟花苷的含量测定。
OBJECTIVE:To establish the method for the content determination of nicotiflorin in Tricyrtis maculate from Shaanxi. METHODS:HPLC method was adopted. The determination was performed on Thermo Hypersil GOLD column with mobile phase consisted of acetonitrile-0.4%H3PO4water solution(gradient elution)at the flow rate of 1 ml/min. The column temperature was set at 20 ℃,and the detection wavelength was 360 nm. RESULTS:The linear range of nicotiflorin was 5.9-35.3 μg(r=0.999 5)with an average recovery of 98.9%(RSD=1.6%,n=6). RSDs of precision,reproducibility and stability tests were less than 2%. CONCLUSIONS:The method is feasible and stable. It can be used for the content determination of nicotiflorin water extract and flavonoids site in T. maculate form Shaanxi.
