摘要
目的:探讨同种异体骨髓单个核细胞(BMMNCs)移植对脊髓全横断损伤区局部微环境中髓鞘碱性蛋白表达的影响。方法:取8周龄Wistar大鼠45只,雌雄不限、每只体重200~220g,随机分为3组:实验组(n=15)、对照组(n=15)、假手术组(n=15)。对实验组和对照组大鼠建立脊髓全横断损伤动物模型。实验组造模后对脊髓横断处上下两端立即注入骨髓单个核细胞;对照组对脊髓横断处上下两端注入磷酸盐缓冲液(PBS);假手术组不损伤脊髓,仅剪除T8~10棘突和椎板后逐层缝合。于细胞移植后的1d,5d,1w,2w,4w和6w分别行BBB大鼠后肢功能评估来判断脊髓神经功能恢复情况,并采用免疫组织化学检测观察大鼠脊髓髓鞘碱性蛋白的表达。结果:(1)BBB评分结果显示各时间点假手术组均为正常范围,实验组和对照组分值随时间均逐渐增加,且实验组较对照组高(P〈0.05),但均未达到正常组水平,术后4周实验组BBB评分较对照组增加显著,差异有统计学意义(P〈0.05);(2)免疫组织染色结果显示各时间点实验组和对照组大鼠脊髓中髓鞘碱性蛋白的表达量高于假手术组,且实验组高于对照组(P〈0.05)。结论:同种异体的骨髓单个核细胞移植可能改变了局部的微环境并上调了脊髓内髓鞘碱性蛋白的表达。
Objective: To investigate the effects of the transplantation of allogeneic bone marrow mononuclear cells (BMMNCs) on myelin basic protein expression in spinal cord injury district local microenvironment. Meth- ods: 45 Winstar rats of either gender aged 8 weeks old, each weighing from 200 to 220g, were randomly divided into three groups: the experimental group (n = 15), the control group (n = 15 ), the sham group (n = 15 ). Rats in the experimental and control groups were established spinal cord injury model. After modeling rats in experimen- tal group were immediately injected into the bone marrow mononuclear cells at the upper and lower ends of the spi- nal cord. Rats in the control group were injected phosphate buffer (PBS) at the upper and lower ends of the spinal cord. The spinal cords in sham group were not damaged. The spines were cut off only T8 - 10 sutured after the sudden and lamina. After the cells were transplanted, BBB hindlimb function in rats was assessed in l d, 5d, l w, 2w, 4w and 6w to determine the recovery of neurological function. Immunohistochemical expression method was used to observe rat spinal cord myelin basic protein. Results: (1) BBB scores showed rats in sham group at each time point were in normal range. Scores in the experimental group and control group were gradually increased with time, and the score in experimental group was higher in the control group (P 〈 0.05 ), but did not reach normal levels. After four weeks, BBB scores in the experimental group increased more significantly than that in the control group, and the difference was statistically significant (P 〈 0. 05). (2) immunohistochemical staining showed that rat spinal cord at each time point in the experimental group and model control group, the expressions of myelin bas- ic protein are higher than those in the sham group; and the experimental group was higher (P 〈 0. 05 ). Conclu- sion : Allogeneic bone marrow mononuclear cell transplantation may change the local microenvironment and raise its expression in the spinal cord myelin basic protein.
