摘要
牛β-乳球蛋白(β-lactoglobulin,BLG)启动子控制人生长激素(h GH)基因的表达载体p BLGH62可以在转基因小鼠的乳腺上皮细胞中高效表达.为了探索利用此表达载体在培养的细胞中生产人生长激素的可能性,将p BLGH62转染非乳腺细胞-人胚肾293细胞用胰岛素、地塞米松和催乳素进行诱导.结果显示,在细胞培养液中检测到人生长激素,说明p BLGH62能在293细胞中表达并能分泌到细胞外.但是,激素诱导对表达量的影响不明显.此外,人生长激素基因存在着可变剪接现象.
Human growth hormone (hGH) gene driven by bovine beta-lactoglobulin (BLG) promoter in vector pBLGH62 was highly expressed in mammary epithelial cells of transgenic mice. To explore the possibility that the production of hGH would be available in cultured cells, the plasmid pBLGH62 was transfected into embryonic kidney 293 cells for transient expression in the presence or absence of induction by insulin, dexamethasoue and prolactin. The results showed that hGH was detected in supernatant, demonstrating that hGH gene expressed in 293 cells and the product secreted outside. However, the effect of hormone induction was compromised. In addition, alternative splicing was observed in the process of hGH gene expression in the cells.
