摘要
目的:观察复方排毒止带汤含药血清对人宫颈鳞癌HPV16 Si Ha细胞E6、E7、p53、Rb m RNA表达的影响。探讨复方排毒止带汤对人乳头瘤病毒(HPV)的影响。方法:将大鼠随机分为空白对照组、25%含药血清组及50%含药血清组。空白对照组予生理盐水灌胃,25%含药血清组及50%含药血清组予复方排毒止带汤煎剂灌胃,制备复方排毒止带汤含药血清,作用于Si Ha细胞,提取总RNA,逆转录为c DNA,进行PCR扩增,PCR产物在2%的琼脂糖凝胶上电泳,采集图像分析结果。结果:复方排毒止带汤50%含药血清作用72 h,HPV16 E6、E7 m RNA表达水平下降,p53、Rb m RNA表达水平上升;复方排毒止带汤50%含药血清作用120 h,HPV16 E6、E7 m RNA表达水平较作用72 h明显下降,p53、Rb m RNA表达水平明显升高,差异均有统计学意义(P<0.05)。复方排毒止带汤25%含药血清作用120 h,HPV16 E6、E7 m RNA表达水平下降,p53、Rb m RNA表达水平上升,复方排毒止带汤50%含药血清作用120 h HPV16 E6、E7 m RNA表达水平较25%含药血清明显下降,p53、Rb m RNA表达水平明显升高,差异均有统计学意义(P<0.05)。结论:复方排毒止带汤含药血清对HPV16 E6、E7、p53、Rb m RNA的表达具有明显调控作用。
Objective: To observe the effect of Fufangpaiduzhidaitang (复方排毒止带汤,,FFPDZDT) drug-containing serum on human cervical carcinoma SiHa cells of human papilloma virus type 16 E6, E7, p53 and Rb mRNA expression, To investigate the effect of FFPDZDT on human papilloma virus (HPV). Methods: The rats are randomly divided into control group, 25% drug-containing serum group and 50% drug-containing serum group. Control group got oral saline solution; the 25% drug- containing serum group and 50% drug-containing serum group got oral FFPDZDT, to prepare medicated serum FFPDZDT drug- containing serum, which played role in SiHa cells. With extraction of total RNA, reverse transcription of eDNA and PCR amplification, the product of PCR was eleetrophoresed on a 2% agarose gel and analysis results. Statistical analysis was performed using independent sample t test. Results: The 50% medicated serum FFPDZDT with 72 hours, HPV16 E6, E7 mRNA expression level decreased and p53 and Rb mRNA expression levels rinsed. But the 50% medicated serum FFPDZDT with 120 hours,comparing with 72 h, the lower HPV16 E6,E7 mRNA expression level decreased and the higher p53 and Rb mRNA expression levels rinsed, with significant difference (P〈0.05) . The 25% medicated serum FFPDZDT with 120 hours, HPV16 E6, E7 mRNA expression level decreased, p53 and Rb mRNA expression levels rinsed. But the 50% medicated serum FFPDZDT with 120 hours, comparing with 25%, the lower HPV16 E6,E7 mRNA expression level decreased and the higher p53 and Rb mRNA expression levels rinsed, with significant difference (P〈0.05). Conclusion: The medicated serum FFPDZDT can provode an obvious regulation effect on the expression of HPV16 E6, E7, p53, Rb mRNA.
