摘要
采用热处理法从海南省尖峰岭热带雨林土壤中分离出164株芽胞杆菌,运用16S rDNA PCR-RFLP与序列分析技术对所得菌株进行遗传多样性分析。根据16S rDNA PCR-RFLP酶切图谱的UPGMA聚类分析,在相似性为100%的水平上,分离到的164株芽胞杆菌分属于23个酶切类型,显示出丰富的遗传多样性。16S rDNA序列分析结果表明,这些菌株主要分布于Bacillus(52%)、Paenibacillus(36%)、Lysinibacillus(4%)、Brevibacillus(4%)和Psychrobacillus(4%)5个属,其中优势属为Bacillus;有6株芽胞杆菌的16S rDNA序列与数据库中相应模式菌株的最大相似性为98.3%~99.0%,可能为潜在的芽胞杆菌新种资源。
A total of 164 Bacillus-like bacterial strains were isolated from Jianfengling tropical rain forest soil through heat-shock treatment.The geneticdiversity of the 164 isolates was analyzed by 16 S rDNA PCR-RFLP and sequence analysis.At the level of 100% similarity,cluster analysis of UPGMA map and 16 S rDNA PCR-RFLP macrorestriction map showed that the isolates were grouped into 23 clusters,which suggested high geneticdiversity.16 S rDNA sequence analysis of representative strains of 23 clusters indicated that there were five genera,Bacillus(52%),Paenibacillus(36%),Lysinibacillus(4%),Brevibacillus(4%) and Psychrobacillus(4%),in which Bacillus was thedominant group.The highest similarities of six strains with the corresponding type strains ranged from 98.3% to 99.0%,which suggested that they might be potential new bacterial species.
