摘要
目的探讨矽肺患者结核易感相关基因的多态性,研究结核易感相关基因对矽肺患者结核发生的影响。方法选择矽肺患者20例,矽肺合并结核患者20例,单纯肺结核患者10例,另选10位健康人作为对照,PCR扩增目的基因,用限制性内切酶对NRAMPl基因3’UTR多态性位点和VDR基因Fok I位点、Taq I多态性位点进行基因分型,各基因型与结核易感性关系用单因素分析和多因素logistic回归分析。结果多因素logistic回归分析表明NRAMP1基因3’UTR和VDR基因Fok I和Taq I位点的多态性与矽肺患者结核发生具有相关性。矽肺合并结核组NRAMP1 3’UTR中TGTG+-/++基因型的频率、TGTG--/++基因型的频率、VDR Fok I和VDR Taq I基因型频率与其它三组相比均升高,差异均有统计学意义(P<0.05)。结论矽肺合并结核患者的NRAMP1基因3’UTR和VDR基因Fok I和Taq I位点的多态性与结核感染存在相关性,NRAMP1基因3’UTR和VDR基因在矽肺合并结核患者的肺结核发病中所起的真正作用以及与其它易感基因或与环境之间的交互作用还需进一步研究确定。
Objective To study the patients with silicosis TB susceptibility genes polymorphisms and susceptibility to tuberculosis related gene in patients with silicosis TB occurs. Methods Twenty cases of silicosis patients,20 patients with silicosis TB merger,10 cases of simple tuberculosis patients,10 healthy people as control were selected for the study,PCR amplification,gene by restriction enzymes of NRAMPl gene 3 ’UTR Fok I system and VDR gene loci polymorphism loci,genotyping Taq I polymorphism loci,the relationship between genotypes and susceptibility to tuberculosis with single factor analysis and multiple factors logistic regression analysis. Results Multi- factor logistic regression analysis showed that 3 ’UTR NRAMPI gene system and VDR Fok I and Taq I polymorphisms of loci associated with patients with silicosis TB occurs. NRAMPI 3 ’UTR gene system and VDR Fok I and Taq I loci polymorphism with silicosis TB susceptibility studies show that compared with the control group,silicosis with tuberculous group,patients with silicosis group,the difference had statistical significance( P 〈 0. 05). Conclusions There was a correlation between patients with silicosis with tuberculous NRAMPI 3 ’UTR gene system and VDR Fok I and Taq I loci polymorphism with tuberculosis,3’ UTR NRAMPI gene system and VDR gene in patients with silicosis with tuberculous played a real role in tuberculosis incidence as well as with other susceptibility genes or the interaction between environment and still need further study.
