摘要
分别以梵净山七叶一枝花种子、根茎、叶片等为实验材料,研究了不同浓度的消毒剂和消毒时间对七叶一枝花组织培养过程中无菌苗获得的影响,通过不同浓度的外源激素(NAA、6-BA、IAA)试验,对七叶一枝花根茎诱导。结果表明:浓度为0.2%的Hg Cl2,消毒8min为七叶一枝花组培消毒的最佳条件,存活率可达50%;启动培养基和愈伤组织诱导培养基分别为MS+1.0mg/L 6-BA+1.0mg/L IAA和MS+1.0mg/L6-BA+0.1mg/L NAA。
Taking rhizome, seeds and leaves of Paris polyphylla as explant, different disinfectants and sterilization times were studied for getting aseptic seedling. Rhizome of P. polyphylla was cultivated in medium with different concentration hormones .The result shows that the best sterilizing condition was 0.2%HgCl2 with 8min, and the sterilizing rate reached 50%; The starter culture and callus induction medium were MS additived 1.0mg/L 6-BA and 1.0mg/L IAA and MS additived 1.0mg/L 6-BA and 0.1mg/L NAA, respectively.
