摘要
目的建立高效液相色谱法(HPLC)测定蛹虫草中4种核苷类物质含量的方法 ,为蛹虫草的开发利用及其产品的指纹图谱提供依据。方法采用Agilent C18色谱柱;以甲醇-水为流动相,采用梯度洗脱,流速为0.8 ml/min,检测波长为260 nm,柱温为30℃。结果样品中尿苷、鸟苷、腺苷和虫草素的加标回收率分别为94.10%、92.23%、94.97%、103.23%,相对标准偏差(RSD)分别为3.75%、5.40%、1.09%、0.30%。结论该法能简单、快速、准确地测定酶解液中4种核苷的含量。
Objective To establish the method of high performance liquid chromatography(HPLC) determining the content of four kinds of nucleotide in Cordyceps militaris to provide the basis for the development and utilization of the fingerprints of Cordyceps militaris and its products. Methods Agilent C18 chromatographic column was used.The mobile phase consisted of methanol-water,and gradient elution was used.The flow rate was 0.8 ml/min,the detection wavelength was 260 nm,the column temperature was 30℃. Results The standard addition recovery rate was 94.10%,92.23%,94.97% and 103.23% respectively,relative standard deviation(RSD) was 3.75%,5.40%,1.09%, 5.40% respectively of uridine,and guanosine,adenosine and cordycep for the sample. Conclusion This method can determine simply,rapidly and accurately the content of 4 kinds of nucleotide in enzymolysis liquid.
