摘要
目的:检测活性氧调节因子1( reactive oxygen species modulator 1,Romo1)在胃癌组织及外周血中的表达,分析其与胃癌临床病理特征的相关性及其意义。方法采用免疫组化S-P法检测40例胃癌患者癌组织及相应的癌旁组织(距癌组织边缘>5 cm)中Romo1蛋白表达;同时用酶联免疫吸附( ELISA)法检测40例胃癌患者及40例健康对照者外周血中Romo1蛋白浓度。结果免疫组化结果显示,胃癌组织中Romo1蛋白阳性表达率(72.5%)显著高于癌旁组织(17.5%)(P<0.01)。胃癌患者外周血中Romo1蛋白浓度为(289.0±51.3)ng/L,显著高于对照组(75.2±18.9)ng/L(P<0.01)。癌组织中Romo1蛋白表达及外周血中Romo1蛋白浓度均与肿瘤浸润深度、TNM分期、淋巴结转移相关(P<0.05或P<0.01),胃癌患者癌组织与外周血中Romo1蛋白表达呈正相关(r=0.774,P<0.01)。结论 Romo1在胃癌患者癌组织及外周血中高表达,其表达与肿瘤浸润深度、TNM分期、淋巴结转移有关,Romo1可能在胃癌的发生过程中起到重要作用;外周血中Romo1浓度能够可靠地反映癌组织中Romo1蛋白表达情况,可作为一种用于胃癌诊断、病情监测的新肿瘤标志物。
Objective To examine the amounts of reactive oxygen species modulator 1 ( Romo1) in the neoplastic tissues and peripheral blood of gastric cancer patients , and to analyze its relationship with the pathological characteristics of gastric cancer .Methods The levels of Romo1 were determined using the neoplastic tissues and corresponding adja-cent normal tissues (〉5 cm from cancer edge ) collected from 40 gastric cancer patients through immunohistochemistry . Meanwhile, the amounts of Romo1 within the peripheral blood of 40 gastric cancer patients and 40 healthy subjects were measured using enzyme -linked immunosorbent assay ( ELISA ) .Results According to immunohistochemistrical re-sults, gastric cancer tissues showed a significant increase in the positive rate of Romol (72.5%) than adjacent normal ones (17.5%) (P〈0.01).The level of Romo1 was measured as (289.0 ±51.3) ng/L in the peripheral blood of gas-tric cancer patients, which were remarkably higher than that of healthy subjects (75.2 ±18.9) ng/L (P〈0.01).The expressions of Romo1 in the neoplastic tissues and peripheral blood were correlated with the depth of infiltration , TNM stage, and lymph node metastasis (P〈0.05 or P〈0.01).The amount of Romo1 in the neoplastic tissues was positively correlated with that in the peripheral blood (r=0.774, P〈0.01).Conclusion The levels of Romo1 were enhanced in the neoplastic tissues and peripheral blood of gastric cancer patients , which may be related with the depth of infiltra-tion, TNM stage and lymph node metastasis .The concentration of Romo 1 within the peripheral blood can reflect the ex-pression of this protein within the neoplastic tissues , which indicates the role of Romol as a new tumor marker for better diagnosing and monitoring of gastric cacner .
